miR-3122靶向CMTM5调控口腔鳞癌细胞GNM凋亡的分子机制 被引量:1

Molecular Mechanism of miR-3122 Targeting CMTM5 to Regulate GNM Apoptosis inOral Squamous Cell Carcinoma Cells

摘要 目的探讨miR-3122靶向CMTM5调控口腔鳞癌细胞GNM凋亡的分子机制。方法纳入口腔鳞癌患者60例,收集其口腔鳞癌组织和癌旁组织,通过实时荧光定量PCR检测miR-3122的表达水平。在口腔鳞癌细胞GNM中敲低或过表达miR-3122,检测其凋亡水平。通过TargetScan,miRcode,miRDB在线分析miR-3122的潜在靶标,通过过表达miR-3122后检测潜在靶标的表达水平。过表达miR-3122的潜在靶标,分析口腔鳞癌细胞GNM的凋亡水平。结果miR-3122在癌旁组织中的表达水平高于口腔鳞癌组织(P<0.05)。过表达miR-3122后,口腔鳞癌细胞GNM的凋亡水平上升(P<0.05),而敲低miR-3122后,口腔鳞癌细胞GNM的凋亡水平下降(P<0.05)。TargetScan,miRcode,miRDB在线分析发现miR-3122分别潜在靶向69,82,184个基因,三者交集为22个基因。过表达miR-3122后,口腔鳞癌细胞GNM中GCSAM,PSMC4,SKP1,LIAS,SMAD2,ZNF704,CMTM5,STK17A,YWHAQ的表达水平下降(P<0.05)。敲低上述基因后,敲低CMTM5后口腔鳞癌细胞GNM的凋亡水平上升(P<0.05)。此外,miR-3122靶向CMTM5的3端非编码区(P<0.05)。过表达miR-3122后,CMTM5的mRNA水平和蛋白水平均下降(P<0.05),而敲低miR-3122后,CMTM5的mRNA水平和蛋白水平均上升(P<0.05)。同时敲低miR-3122和CMTM5时,口腔鳞癌细胞GNM凋亡水平无显著变化(P<0.05);同时过表达miR-3122和CMTM5时,口腔鳞癌细胞GNM凋亡水平无显著变化(P<0.05)。结论miR-3122通过靶向CMTM5 mRNA的3端非编码区后,降解了CMTM5的mRNA并降低了CMTM5的蛋白水平,随后促进了口腔鳞癌细胞GNM的凋亡。 Objective To investigate the molecular mechanism of miR-3122 targeting CMTM5 to regulate GNM apoptosis in oral squamous cell carcinoma cells.Methods 60 patients with oral squamous cell carcinoma were enrolled.The oral squamous cell carcinoma tissues and paracancerous tissues were collected,and the expression level of miR-3122 was detected by real-time fluorescent quantitative PCR.Knock down or overexpress miR-3122 in oral squamous cell carcinoma cell GNM,and detect its apoptosis level.Through TargetScan,miRcode,miRDB online analysis of potential targets of miR-3122,through overexpression of miR-3122 to detect the expression level of potential targets.Overexpress the potential targets of miR-3122,and analyze the apoptosis level of oral squamous cell carcinoma cell GNM.Results The expression level of miR-3122 in adjacent tissues was higher than that in oral squamous cell carcinoma tissues(P<0.05).After overexpression of miR-3122,the apoptotic level of oral squamous cell carcinoma cell GNM increased(P<0.05),and after knocking down miR-3122,the apoptotic level of oral squamous cell carcinoma cell GNM decreased(P<0.05).Online analysis of TargetScan,miRcode,and miRDB found that miR-3122 potentially targets 69,82,and 184 genes,respectively,and the intersection of the three was 22 genes.After overexpression of miR-3122,the expression levels of GCSAM,PSMC4,SKP1,LIAS,SMAD2,ZNF704,CMTM5,STK17A,and YWHAQ in oral squamous cell carcinoma cells GNM decreased(P<0.05).After knocking down the above genes,the apoptosis level of oral squamous cell carcinoma cell GNM increased after knocking down CMTM5(P<0.05).In addition,miR-3122 targets the 3'non-coding region of CMTM5(P<0.05).After overexpression of miR-3122,the mRNA level and protein level of CMTM5 decreased(P<0.05),while after knocking down miR-3122,the mRNA level and protein level of CMTM5 increased(P<0.05).When miR-3122 and CMTM5 wereknocked down at the same time,the GNM apoptosis level of oral squamous cell carcinoma cells did not change significantly(P<0.05);when miR-3122 an
