摘要
目的采用网络药理学方法探讨生脉饮"异病同治"糖尿病和心力衰竭的作用机制。方法通过中药系统药理学数据库与分析平台(TCMSP)和BATMAN-TCM数据库收集生脉饮组方药物的所有化学成分,通过UniProt数据库获取药物作用靶点,通过DisGeNET和GeneCards数据库筛选与糖尿病和心力衰竭相关的靶点,将药物有效成分作用靶点与疾病靶点比对筛选后,分别构建生脉饮治疗糖尿病和心力衰竭的化合物-靶点相互作用网络图;利用String平台绘制蛋白相互作用(PPI)网络,找出关键基因靶点;分别使用DAVID数据库和Cytoscape3.6.1对核心靶点进行GO功能富集和KEGG通路富集,并分析PPI网络数据。结果共筛选出生脉饮50个活性成分和3754个潜在作用靶点,糖尿病、心力衰竭相关靶点分别为218、317个,生脉饮治疗糖尿病、心力衰竭作用核心靶点分别为109、196个;经KEGG通路富集分析,分别筛选出生脉饮对糖尿病和心力衰竭具有作用的通路,根据度值对PPI网络数据分析选取生脉饮治疗糖尿病和心力衰竭的主要药效靶点各10个。结论生脉饮治疗糖尿病和心力衰竭具有多成分、多靶点、整体调节的作用特点,可为其活性成分研究和实验研究提供依据。
Objective To explore the mechanism of treating different diseases with same method of Shengmai Decoction in treating diabetes and heart failure by network pharmacology.Methods After searching for all the chemical components related to the medicine in Shengmai Decoction from TCMSP and BATMAN-TCM platform,medicine targets in UniProt database were found,and the targets of diabetes and heart failure were screened through DisGeNET and GeneCards database.Compound-target interaction network maps of Shengmai Decoction for the treatment of diabetes and heart failure were constructed;String platform was used to draw a protein-protein interaction(PPI)network to identify key gene targets;DAVID database and Cytoscape 3.6.1 were used to perform GO function enrichment analysis and KEGG pathway enrichment analysis on core targets,respectively,and PPI network data were analyzed.Results Totally 50 active ingredients and 3754 potential targets of Shengmai Decoction were screened out,218 and 317 targets were related to diabetes and heart failure,respectively,and 109 and 196 core targets were related to treating diabetes and heart failure in Shengmai Decoction.Based on KEGG pathway enrichment analysis,the pathways of Shengmai Decoction for the treatment of diabetes and heart failure were screened out.According to the degree value of the PPI network data analysis,10 main targets of Shengmai Decoction for the treatment of diabetes and heart failure were selected.Conclusion Shengmai Decoction has the characteristics of multi-component,multi-target and overall regulation in the treatment of diabetes and heart failure,which can provide a basis for research on its active ingredient and experimental research.
