摘要
目的观察电针内关穴对心肌缺血大鼠心脏功能及心肌组织中Fbxo32 mRNA表达的影响。方法SD大鼠随机分成生理盐水对照组、模型组和电针组。24 h内大鼠四肢背部皮下2次注射异丙肾上腺素(ISO),造成心肌缺血模型。电针组造模24 h后电针内关穴20 min,连续5 d。生理盐水对照组和模型组予以同等抓取处理。通过心肌酶肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)活性、病理苏木素-伊红(HE)染色观察各组大鼠心脏功能变化;基因芯片检测心肌泛素连接酶E3;实时荧光定量PCR(q-PCR)检测心肌组织Fbxo32 mRNA表达。结果电针组与模型组比较,心肌泛素连接酶E3基因有显著差异表达;模型组心肌酶CK-MB、LDH活性、Fbxo32 mRNA表达量均显著高于生理盐水对照组(P<0.01);电针组心肌酶CK-MB、LDH活性、Fbxo32 mRNA表达量均显著低于模型组(P<0.05)。结论电针可改善心肌缺血大鼠心脏功能,其机制可能与下调心肌组织中Fbxo32表达有关。
Objective To observe the effects of electro-acupuncture(EA)on cardiac function and the expression of Fbxo32 mRNA in the myocardial tissue of myocardial ischemia rats.Methods SD rats were randomly divided into three groups:control group,myocardial ischemia model group and EA group.Myocardial infarction was produced in rats with 85 mg/kg of isoproterenol administered subcutaneously twice at an interval of 24 h.Rats of EA group were treated with EA at Neiguan Point(PC6)for 5 days.Rats of control group and model group were captured under the same conditions.The activity of CK-MB and LDH,HE staining were observed as indicators of cardiac function.The myocardial tissue was taken for gene chip to detect the expression of E3 ubiquitin ligases.Real-time quantitative fluorescent PCR was used to detect the expression of Fbxo32 mRNA in the myocardial tissue.Results Compared with the model group,the expression of myocardial E3 ubiquitin ligase genes were significantly different in EA group.The activity of CK-MB and LDH,Fbxo32 mRNA expression in the model group were significantly higher than those in the control group(P<0.01,P<0.05,P<0.01).While those in the EA group were significantly lower than those of model group(P<0.05).Conclusions EA could improve cardiac function in myocardial ischemia rats,and its mechanism might be related to the down-regulation of Fbxo32 expression in the myocardial tissue.
