预电针调节胆碱能神经相关蛋白对AD样大鼠学习记忆能力及脑内炎症的影响 被引量:15

Effect of pre-electro-acupuncture on cholinergic nerve-related proteins on learning and memory abilities and inflammation in the brain of AD-like rats

摘要 目的观察预电针对AD样大鼠学习记忆能力及脑内炎症的影响。方法30只SD大鼠分成正常组、模型组、预电针组,每组10只。正常组不做干预,模型组、预电针组每日腹腔注射D半乳糖溶液制备AD样大鼠模型,预电针组另予以针刺百会、足三里穴,接HANS-200A韩式电针仪,连续波,电流1 mA,频率50 Hz,1次/d,20 min/次,连续治疗8 w,造模与治疗同步。实验结束后,采用Morris水迷宫检测大鼠行为学,免疫荧光法检测海马小胶质细胞离子钙接头蛋白(Iba)-1的表达,免疫组织化学法检测海马区胆碱乙酰转移酶(ChAT)的表达,Western印迹法检测大鼠海马组织α7烟碱受体(α7nAChR)、Iba-1的表达,酶联免疫吸附试验(ELISA)检测肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β、IL-6的含量。结果与正常组相比,模型组α7nAChR、ChAT表达显著降低(P<0.01),Iba-1、TNF-α、IL-1β、IL-6表达显著增多(P<0.01);与模型组相比,预电针组α7nAChR、ChAT表达显著增多(P<0.05),Iba-1、TNF-α、IL-1β、IL-6表达显著降低(P<0.05)。结论预电针能改善AD样大鼠学习记忆能力及脑内炎症反应,其机制可能与预电针调节胆碱能神经相关蛋白有关。 Objective To observe the effect of pre-electro-acupuncture preconditioning on learning and memory ability and inflammation in brain of AD-like rats.Methods Thirty SD rats were divided into normal group,model group and pre-electro-acupuncture group,with 10 rats in each group.The normal group did not intervene.The model group and the pre-electro-acupuncture group were injected intraperitoneally with D-galactose solution to prepare AD-like rat models.The pre-electro-acupuncture group was additionally given acupuncture points at"Baihui"and"Zusanli"and connected to HANS-200 A Korean electro-acupuncture instrument(continuous wave,current 1 mA,frequency 50 Hz,once a day,20 min/time),continuous treatment last for 8 weeks,modeling and treatment were synchronized.After the experiment,Morris water maze was used to detect the behavior of rats.The expression of hippocampal microglia ionized calcium binding adapter molecule(Iba)-1 was detected by immunofluorescence.The expression of choline acetyltransferase(ChAT)in hippocampus was detected by immunohistochemistry.The expression ofα7 nicotinic receptor(nAChR)and Iba-1 in rat hippocampus were detected by Western blot.The levels of tumor necrosis factor(TNF)-α,interleukin(IL)-1βand IL-6 were detected by ELISA.Results Compared with the normal group,the expressions ofα7 nAChR and ChAT were significantly decreased(P<0.01),and the expressions of Iba-1,TNF-α,IL-1βand IL-6 were significantly increased in the model group(P<0.01).Compared with the model group,the expressions ofα7 nAChR and ChAT were significantly increased(P<0.05),and the expressions of Iba-1,TNF-α,IL-1βand IL-6 were significantly decreased in the electro-acupuncture group(P<0.05).Conclusions Pre-electro-acupuncture could improve the learning and memory ability and brain inflammatory response in AD-like rats,and its mechanism might be related to pre-electro-acupuncture regulating cholinergic nerve-associated protein.
