摘要
目的研究补阳还五汤对脑缺血后大鼠LAMP2蛋白表达的影响。方法2019年7月16日—2019年7月30日用改良线栓法制备大鼠中动脉栓塞模型,将50只大鼠随机分成5组,以补阳还五汤不同浓度进行灌胃治疗,每天灌胃1次,造模前灌胃7天,造模后继续灌胃7天。取造模后大鼠脑细胞用Western blot法进行LAMP2蛋白表达测定。探讨不同浓度补阳还五汤对LAMP2蛋白表达的影响。结果假手术组LAMP2蛋白表达水平明显高于模型组、常规、2倍及10倍剂量组,差异有统计学意义(P<0.01)。模型组LAMP2蛋白表达水平明显低于假手术组、2倍及10倍剂量组(P<0.01)。假手术组LAMP2表达水平最高,模型组最低,模型组与假手术组、2倍剂量组、10倍剂量组比较,差异有统计学意义(P<0.01),与常规剂量组比较,差异无统计学意义(P>0.05)。结论补阳还五汤常规剂量治疗基本无效,10倍剂量组对脑缺血后大鼠的治疗效果确切有效。补阳还五汤可能通过上调LAMP2蛋白的表达使CMA活性上调进而发挥神经保护作用。
Objective To study the effect of Buyang Huanwu decoction on the expression of LAMP2 protein in rats after cerebral ischemia.Methods Middle artery embolization model was established in rats by improved suture method from July 16,2019 to July 30,2019,50 rats were randomly divided into 5 groups.The rats were given Buyang Huanwu decoction with different concentrations by gavage once a day,7 days before modeling and 7 days after modeling.The expression of LAMP2 protein was detected by Western blot.Objective to investigate the effect of Buyang Huanwu decoction on LAMP2 protein expression.Results The expression level of LAMP2 protein in sham operation group was significantly higher than that in model group,routine group,2-fold and 10 fold dose group(P<0.01).The expression level of LAMP2 protein in model group was significantly lower than that in sham operation group,2-fold and 10 fold dose group(P<0.01).The expression level of LAMP2 was the highest in the sham operation group and the lowest in the model group.There was significant difference between the model group and the sham operation group,the double dose group and the 10 times dose group(P<0.01).There was no significant difference between the model group and the conventional dose group(P>0.05).Conclusion Routine dose treatment of Buyang Huanwu decoction is basically ineffective,the therapeutic effect of Buyang Huanwu decoction 10 times dose group on rats with cerebral ischemia is effective.Buyang Huanwu decoction may play a neuroprotective role by up regulating the expression of LAMP2 protein.
