云南文山市石漠化土壤真菌多样性研究 被引量:2

Diversity Research of Soil Fungi in Rocky Desertification Area of Wenshan City

摘要 为阐明文山市石漠化土壤真菌多样性情况及优势关键类群,采集石漠化样区中植物根际土壤,提取土壤总DNA,利用Illumina HiSeq平台对真核微生物ITS(Internal Transcribed Spacer)区扩增进行高通量测序后注释鉴定,分析了土壤真菌组成及多样性。结果表明,典型样本共获得168677条有效优化序列,平均长度为452.7 bp;α多样性指数显示3个样本间的真核微生物丰度与群落多样性差异不显著(P>0.05);检测到的序列聚类共得到314个OTUs,属于真菌的有7门,其中子囊菌门是占比最高的一门优势菌,其相对丰度最高值为71.07%,从纲阶元层看,座囊菌纲是占比最高的一属优势菌,其相对丰度最高值为37.96%;在目阶元层看格孢腔目是占比最高的一目优势菌,范围在20.67%~21.71%、其次是煤炱目(14.29%~15.66%)和肉座菌目(19.24%~20.38%);从属水平上看未分类真核微生物达52.70%~53.98%,其次是Boeremia(15.10%~16.88%)和芽枝霉属13.25%~14.28%)、旋孢腔菌属(3.66%~6.12%)、丝核菌属(2.70%~3.24%)、伞菌属(2.53%~2.87%)、球腔菌属(1.04%~1.43%)组成了关键优势真菌类群;样本中仅鉴定出球囊霉目的AM真菌,占比较低为1.05%~1.09%。Heatmap热图和主成分分析(PCA)显示样本群落结构组间差异较大。 To illustrate the diversity of soil fungal in rocky desertification area of Wenshan city and find out the predominant groups,the samples are collected from rhizosphere soil of plants in rocky desertification-like areas,soil total DNA is extracted,and paper uses the Illumina HiSeq platform on ITS eukaryotic microbes(Internal Transcribed Spacer)to determine after the high-throughput sequencing identification of area augmentation,and analyzes the composition of soil fungi and diversity.Results show that:a total of 168677 effective and majorized sequences from the soils typical samples are obtained,the average length is 452.7bp.Theαdiversities index indicate undiffered significantly in three samples(P>0.05).A total of 314 OTUs are detected,belonging to 7 fungi plylas,among which Ascomycota of dominant microflora account for a relatively high proportion which is up to 69.23~71.07%.At class level,the dominant microflora,Dothideomycetes and Sordariomycetes,account for a relatively high proportion,with relative abundance of 36.67~37.96%and 20.22~21.50%,respectively.At order level,the dominant microflora are Platyophryid(20.67~21.71%),Capnodiales(14.29~15.66%)and Hypocreales(19.24~20.38%).At generic level,the unclassified eukaryotic microorganisms are up to 52.70~53.98%,followed by Boeremia(15.10~16.88%),Cladosporium(13.25~14.28%),Cochliobolus(3.66~6.12%),Rhizoctonia(2.70~3.24%),Agaricus(2.53~2.87%)and Mycosphaerella(1.04~1.43%).In the samples,only AM fungi of Glomerales were identified,accounting for 1.05~1.09%.The Heatmap and principal component analysis(PCA)shows that there are significant differences in the sample community structure between groups.
