摘要
穇子作为药食兼用的特色作物,对粮食安全和改善人们健康起着重要作用。为探究快速鉴定穇属物种的方法及确定穇子的亲缘关系,本研究对65个不同产地穇子居群的核糖体DNA内转录间隔区(ITS)序列进行测序及比对,并构建穇属物种系统发育树。结果发现,穇子EC25、EC53和EC59等3个居群的ITS序列存在单碱基的突变体,其他居群无任何差异;穇属物种间ITS存在3~43个碱基的差异;系统聚类表明,穇属物种分为2大类4个亚组,其中穇子与凯芝济穇亲缘关系最近。研究表明,穇子的ITS序列具有高保守性而穇属种间差异性较大,ITS序列不能作为穇子居群的鉴定标记,可作为穇属物种鉴定的参考序列。本研究为穇子的亲缘关系鉴定及杂交育种提供了分子依据。
Finger millet,as a special crop for both medicine and food,plays an important role in food security and improves people's health.In order to establish a method for rapid identification of finger millet(Eleusine coracana)and determine the genetic relationship of finger millet,the ribosomal DNA transcribed spacer(ITS)sequences of 65 finger millet populations from different regions were sequenced and compared,and the phylogenetic tree of Eleusine was constructed.It was found that there were single base mutants in the ITS sequences of the three finger millet populations,including EC25,EC53,and EC59,and there was no difference in other populations.There are 3 to 43 base differences in ITS between Eleusine species.The phylogenetic tree showed that the Eleusine species are divided into 2 subgroups of 4 categories,of which finger millet has the highest phylogenetic relationship with Eleusine kigeziensis.Studies have shown that the ITS sequences of finger millet are highly conserved,but ITS is highly different among Eleusine.This indicates that the ITS sequence cannot be used as an identification marker for the population of finger millet,but can be used as a reference sequence for the identification of Eleusine.This study provided molecular basis for the identification of genetic relationship and cross breeding of finger millet.
