赤芍总苷对脊髓损伤大鼠脊髓细胞凋亡的影响及机制 被引量:6

Effect and mechanism of the total paeony glycosides on spinal cord apoptosis in rats with spinal cord injury

摘要 目的探讨赤芍总苷(TPG)对脊髓损伤(SCI)大鼠脊髓细胞凋亡的影响及机制。方法SD大鼠18只,随机分为对照组、SCI组与TPG组,每组6只。采用Allen法制备SCI大鼠模型。对照组大鼠切除椎板后,不予以脊髓打击直接缝合。TPG组在SCI模型建立成功后每24 h腹腔注射25 mg/kg TPG,SCI组和对照组腹腔注射同体积的生理盐水,共处理28 d。采用Basso-Beattie-Bresnahan(BBB)评分法对大鼠进行行为学评分,流式细胞术检测脊髓细胞凋亡情况,Western blotting检测脊髓细胞凋亡蛋白cleaved caspase-3和cleaved caspase-9的表达。采用试剂盒检测脊髓组织中丙二醛(MDA)、超氧化物歧化酶(SOD)和乳酸脱氢酶(LDH)的含量。结果与对照组比较,SCI大鼠BBB评分明显降低,脊髓细胞凋亡率以及凋亡蛋白cleaved caspase-3、cleaved caspase-9表达明显增高,脊髓组织中MDA、LDH含量升高,SOD含量降低(P<0.05)。与SCI组比较,TPG组大鼠BBB评分明显增高,脊髓细胞凋亡率以及凋亡蛋白cleaved caspase-3、cleaved caspase-9表达明显降低,脊髓组织中MDA、LDH含量降低,SOD含量升高(P<0.05)。结论TPG对SCI大鼠脊髓细胞凋亡有抑制作用,其作用机制可能与TPG对氧化应激的调节有关。 Objective To investigate the effect and mechanism of total paeony glycosides(TPG)on spinal cord apoptosis in rats with spinal cord injury(SCI).Methods A total of 18 SD rats were randomly divided into control group,SCI group and TPG group(6 each).The SCI rat model was established using Allen method.For rats in control group,the spinal cord was sutured directly after laminectomy without spinal cord injury;those in TPG group were intraperitoneally injected with 25 mg/kg TPG every 24 hours after the establishment of SCI model,while those in SCI group and control group were intraperitoneally injected with the same volume of normal saline for 28 days.The rats'behavioral score were evaluated using Basso-Beattie-Bresnahan(BBB)method.The apoptosis of spinal cord cells was measured by flow cytometry.Western blotting was performed to detect the expression levels of cell apoptotic protein cleaved caspase-3 and cleaved caspase-9.The contents of MDA,SOD and LDH in the supernatant of spinal fluid were quantified with corresponding kits.Results Compared to those in control group,the BBB score decreased significantly,while the apoptosis rate of spinal cord cells and the expression levels of cleaved caspase-3 and cleaved caspase-9 were obviously elevated,and the contents of MDA and LDH increased,SOD decreased in the supernatant of spinal fluid in the rats of SCI group(P<0.05).Compared to those in SCI group,after treatment with TPG,the BBB score increased significantly,the apoptosis rate of spinal cord cells and the expression levels of cleaved caspase-3 and cleaved caspase-9 decreased obviously,while the contents of MDA and LDH decreased,and SOD increased in the supernatant of spinal fluid of the rats in SCI group(P<0.05).Conclusion TPG inhibits the apoptosis of spinal cord cells in SCI rats,which may related to the function of TPG in regulating oxidative stress.
