miR-5088-5p在子痫前期患者胎盘组织中的表达及其对滋养细胞自噬的影响 被引量:5

摘要 目的探讨胎盘组织miR⁃5088⁃5p表达水平与子痫前期患者临床特征的相关性及其在子痫前期发病机制中的作用。方法qRT⁃PCR检测胎盘组织miR⁃5088⁃5p的表达;将miR⁃5088⁃5p表达水平与临床特征进行相关性分析;免疫荧光检测胎盘组织中LC3B及p62的表达;同时,体外培养人源胎盘滋养细胞株(HTR8⁃S/Vneo),分为常氧组和缺氧组,转染miR⁃5088⁃5p mimics后,Western blot检测各组胎盘滋养细胞LC3B和p62蛋白的表达。结果miR⁃5088⁃5p在子痫前期胎盘组织中明显低表达(P<0.01),且表达水平与收缩压、舒张压、血小板计数呈负相关,而与新生儿体重呈正相关(P<0.05);免疫荧光显示LC3B在子痫前期组胎盘组织中增加,而p62减少;另外,与常氧组比较,缺氧组细胞LC3BⅡ/Ⅰ蛋白表达增加(P<0.05),而p62蛋白表达降低(P<0.05);转染miR⁃5088⁃5p mimics后,LC3BⅡ/Ⅰ表达降低(P<0.01),p62表达增加(P<0.05)。结论miR⁃5088⁃5p在子痫前期胎盘组织中显著下调,其过表达可显著抑制胎盘滋养细胞自噬的发生,有望成为子痫前期的诊断分子标记物和治疗靶点。 Objective To explore the relationship between the expression of miR⁃5088⁃5p in placenta and the clinical characteristics of patients with preeclampsia and its role in the pathogenesis of preeclampsia.Methods The expression of miR⁃5088⁃5p in placenta was measured by qRT⁃PCR and the correlation between the expression of miR⁃5088⁃5p and clinical features was analyzed,and the expression of LC3B and p62 in placental tissue was detected by immunofluorescence.At the same time,human placental trophoblast cell line(HTR8⁃S/Vneo)was cultured in vitro and divided into nomorxia group and hypoxia group.The expression of LC3B and p62 protein in placental trophoblasts of each group was detected by Western blot after transfection of miR⁃5088⁃5p mimics.Results The expression of miR⁃5088⁃5p in placenta of preeclampsia was significantly low(P<0.01),and the expression level was negatively correlated with systolic blood pressure,diastolic blood pressure and platelet count,but positively correlated with neonatal weight(P<0.05).Immunofluorescence showed that LC3B increased and p62 decreased in placenta of preeclampsia group.In addition,compared with the nomorxia group,the expression of LC3BⅡ/Ⅰprotein in hypoxia group increased(P<0.05),while the expression of p62 protein decreased(P<0.05),and the expression of LC3BⅡ/Ⅰdecreased(P<0.01)and p62 increased(P<0.05)after transfection of miR⁃5088⁃5p mimics.Conclusion miR⁃5088⁃5p is significantly down⁃regulated in placental tissue of preeclamp⁃sia,and its overexpression can significantly inhibit the occurrence of autophagy of placental trophoblasts,which is expected to be a diagnostic molecular marker and therapeutic target for preeclampsia.
