荆门上清丸对动物模型的抗炎活性及血清PGE2、IL-1β含量的影响 被引量:3

摘要 目的研究荆门上清丸对大鼠、小鼠的抗炎作用,并探讨其抗炎活性与大鼠血清前列腺素E2(PGE2)、白介素-1β(IL-1β)的关系。方法通过二甲苯致小鼠耳廓肿胀,角叉菜胶致大鼠足跖肿胀实验观察荆门上清丸的抗炎活性。各炎性动物模型随机均分为5组,小鼠模型每组10只,大鼠模型每组8只,各分为对照组(0.2 mL/10 g生理盐水),荆门上清丸高、中、低剂量组(1000、500、250 mg/kg)组,阿司匹林(100 mg/kg)组,每天灌胃给药1次,连续3 d,末次给药1 h后,小鼠右耳均匀涂抹二甲苯溶液30μL致炎,测量小鼠双耳质量差,计算耳廓肿胀抑制率;大鼠右后足掌皮下注射2%角叉菜胶100μL,在注射后1、2、3、4、5 h测定大鼠右后足跖肿胀度,最后取眼球血约1 mL,用ELISA法检测血清中PGE2、IL-1β的含量。结果与对照组比较,荆门上清丸高、中剂量组能抑制二甲苯致小鼠耳廓肿胀(P<0.05),并降低实验大鼠血清中PGE2、IL-1β含量(P<0.05);在注射2%角叉菜胶角后3、4、5 h,与对照组比较,荆门上清丸高、中、低剂量组均可抑制大鼠足跖肿胀(P<0.05)。结论荆门上清丸对急性炎症动物模型表现出良好的抗炎活性,其机制可能与抑制血清PGE2、IL-1β的含量有关。 Objective To study the anti-inflammatory effect of Jingmen Shangqing pill in rats and mice and to further explore the relationships between its anti-inflammatory activity and serum prostaglandin E2(PGE2),interleukin-1β(IL-1β)in rats.Methods The anti-inflammatory activity of Jingmen Shangqing pill was observed by xylene-induced ear swelling in mice and carrageenan-induced toe swelling in rats.The 50 inflammatory mouse models and 40 rat models were randomly divided into 5 groups:control group(0.2 mL/10 g saline),Jingmen Shangqing pill high,medium and low dose groups(1000,500,250 mg/kg)and aspirin(100 mg/kg)group.The mice were given the drug once a day for 3 consecutive days.At 1 h after the last drug was given,the right ear of the mice was uniformly coated with 30μL xylene solution,causing inflammation.The difference in the quality of both ears was measured and the inhibition rate of auricle swelling was calculated.Rats were subcutaneously injected with 2%carrageenan 100μL in the right hind paw,and the swelling degree of right hind foot was determined at 1,2,3,4,and 5 h after the injection.Finally,about 1 mL of eyeball blood was taken.Contents of PGE2 and IL-1βin serum were determined by ELISA.Results Compared with the control group,Jingmen Shangqing pill high and medium dose groups could inhibit xylene-induced auricle swelling in mice(P<0.05)and reduce PGE2 content(P<0.05)and IL-1βcontent in serum of model rats(P<0.05).At 3,4 and 5 h after 2%carrageenan injection,compared with the control group,the swelling of foot plantarum in the high,medium and low dose Jingmen Shangqing pill group could be inhibited(P<0.05).Conclusion Jingmen Shangqing pill has good anti-inflammatory activity on acute inflammatory models,and its mechanism may be related to the inhibition of PGE2 and IL-1βcontents in serum.
