中风1号方对急性脑出血大鼠的保护作用及机制研究 被引量:4

摘要 目的:观察中风1号方对急性脑出血(acute intracerebral hemorrhage,AICH)模型大鼠的影响,探讨其初步机制。方法:采用自体尾部血液注入法复制大鼠急性脑出血模型。将大鼠随机分为假手术组、模型组、中风1号方低剂量(6.75 g·kg^-1)组和中风1号方高剂量(13.5 g·kg^-1)组,连续给予相应的药物灌胃3天。用Longa 5级评分法评估神经功能损伤程度;计算脑组织含水量;检测脑组织伊文思蓝(evans blue,EB)含量,评价血脑屏障通透性;ELISA法检测脑出血周围组织肿瘤坏死因子α(tumor necrosis factor,TNF-α)、白细胞介素1β(interleukin-1β,IL-1β)水平。结果:与假手术组比较,模型组大鼠24 h、72 h的Longa 5级评分,脑组织含水量,EB含量,TNF-α、IL-1β水平均升高(P<0.05,P<0.01)。与模型组比较,中风1号方低剂量可降低模型大鼠神经行为学评分(72 h),脑组织含水量(72 h),EB含量,TNF-α、IL-1β水平(72 h)(P<0.05,P<0.01);中风1号方高剂量组可降低模型大鼠的神经行为学评分(72 h),脑组织含水量(72 h),EB含量(24 h、72 h),TNF-α、IL-1β水平(24 h、72 h)(P<0.01)。结论:中风1号方能改善急性脑出血模型大鼠神经功能损伤,减轻脑水肿,缓解血脑屏障受损,其机制可能与降低脑组织TNF-α、IL-1β水平有关。 Objective:To observe the effect of Zhongfeng No.1 Prescription on acute intracerebral hemorrhage(AICH)model rats and explore its preliminary mechanism.Methods:The model of acute cerebral hemorrhage in rats was replicated by autologous tail blood injection.The rats were randomly divided into sham operation group,model group,Zhongfeng No.1 Prescription low-dose(6.75 g·kg^-1)group and Zhongfeng No.1 Prescription high-dose(13.5 g·kg^-1)group,and given corresponding drugs continuously 3 days.The Longa 5-level scores method was used to evaluate the degree of neurological damage;calculate the water content of brain tissue;detect the content of evans blue(EB)in brain tissue to evaluate the permeability of the blood-brain barrier;the ELISA method was used to detect the levels of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)in tissues surrounding cerebral hemorrhage.Results:Compared with the sham operation group,the Longa 5-level scores,brain tissue water content,EB content,TNF-α,IL-1βlevels of the model group increased at 24 h and 72 h(P<0.05,P<0.01);compared with the model group,the low dose of Zhongfeng No.1 prescription can reduce the water content of brain tissue(72 h),EB content,TNF-α,IL-1βlevels(72 h)(P<0.05,P<0.01);the high-dose group of Zhongfeng No.1 prescription can reduce the neurological function score(72 h),brain tissue water content(72 h),EB content(24 h,72 h),TNF-α,IL of model rats-1βlevel(24 h,72 h)(P<0.01).Conclusion:Zhongfeng No.1 Prescription can improve the nerve function damage of acute cerebral hemorrhage model rats,reduce brain edema,and relieve blood-brain barrier damage.The mechanism may be related to the reduction of brain tissue TNF-αand IL-1βlevels.
