乳腺癌组织的GATA6-AS1水平和临床意义 被引量:2

摘要 目的探讨长链非编码RNA GATA6反义RNA 1(GATA6-AS1)在乳腺癌组织的水平和临床意义。方法采用实时定量PCR(qPCR)检测本院2013年1月至2018年12月手术切除的97对乳腺癌组织和配对癌旁组织的GATA6-AS1水平,比较不同组织的GATA6-AS1水平差异,以乳腺癌组织GATA6-AS1水平的中位值为参考将97例乳腺癌组织分为低水平组和高水平组以进一步分析GATA6-AS1水平与乳腺癌临床病理特征(年龄、肿瘤大小、ER表达、PR表达、HER-2扩增、组织学分级、TNM分期、淋巴结转移和绝经状态)的关系,根据随访数据分析不同GATA6-AS1水平的预后情况并进行多因素分析。结果乳腺癌组织的GATA6-AS1水平为0.512±0.240,低于癌旁组织的1.761±0.739(t=13.054,P<0.001)。以组织GATA6-AS1水平的中位值(0.532)为参考分为低水平(≤0.532)组49例和高水平组(>0.532)组48例,单因素分析发现GATA6-AS1水平与肿瘤大小、TNM分期、淋巴结转移和HER-2扩增有关(P<0.05),而与年龄、组织学分级、ER表达、PR表达和绝经状态无关(P>0.05)。全组97例患者随访11~60个月,中位随访40个月,中位无病生存期(DFS)为51.0个月,其中49例低水平者的中位DFS为45.0个月,短于48例高水平者的54.0个月,差异有统计学意义(P=0.002);进一步多因素分析发现组织GATA6-AS1水平降低是乳腺癌预后不良的独立因素(HR=3.126,95%CI:1.478~5.712,P=0.002)。结论乳腺癌组织的GATA6-AS1水平降低,且与较大肿瘤体积、分期晚、淋巴结转移和HER-2扩增有关,对乳腺癌的诊治指导和预后评价有一定价值。 Objective To investigate the level and clinical significance of long noncoding RNA GATA6 antisense RNA 1(GATA6-AS1)in breast cancer tissues.Methods Real-time quantitative PCR(qPCR)was used to detect the GATA6-AS1 levels in 97 pairs of breast cancer tissues and matched adjacent tissues from January 2013 to December 2018 in our hospital.The differences of GATA6-AS1 levels in different tissues were compared.According to the median value of GATA6-AS1 level in breast cancer tissues,97 cases of breast cancer tissues were divided into low-level group and high-level group,in order to further analyze the relationship between GATA6-AS1 level and clinicopathological features(age,tumor size,ER expression,PR expression,HER-2 amplification,histological grade,TNM stage,lymph node metastasis and menopausal status).The prognosis of different GATA6-AS1 levels was analyzed according to the follow-up data.Results GATA6-AS1 level in breast cancer tissues was 0.512±0.240,which was lower than 1.761±0.739 in adjacent tissues(t=13.054,P<0.001).According to the median value(0.532)of GATA6-AS1 level,patients were divided into low-level(≤0.532)group(49 cases)and high-level(>0.532)group(48 cases).Univariate analysis showed that GATA6-AS1 level was related to tumor size,TNM stage,lymph node metastasis and HER-2 amplification(P<0.05),but not with age,histological grade,ER expression,PR expression and menopausal status(P>0.05).All patients were followed up for 11-60 months with a median follow-up of 40 months and a median disease-free survival(DFS)of 51.0 months.The median DFS of 49 patients with low-level GATA6-AS1 was 45.0 months,which was shorter than 54.0 months of 48 patients with high-level GATA6-AS1(P=0.002).Further multivariate analysis showed that the decrease of GATA6-AS1 level was an independent factor for poor prognosis of breast cancer(HR=3.126,95%CI:1.478~5.712,P=0.002).Conclusion The level of GATA6-AS1 in breast cancer tissue was decreased,which was associated with larger tumor size,advanced stage,lymph node metastasis and HE
