类风湿关节炎患者血清长链非编码RNA表达谱的差异性分析 被引量:4

摘要 目的血清长链非编码RNA与类风湿关节炎(RA)的发展紧密相关。文中旨在探讨RA患者血清内lncRNA的差异性表达。方法收集2017年11月至2018年11月于东部战区总医院(原南京军区南京总医院)中西医结合科入院的RA患者和门诊健康体检人员血清标本。其中RA患者血清标本30份(RA组),健康体检人员的血清标本30份(对照组)。采用基因芯片技术对血清进行LncRNAs差异表达及生物信息学分析。结果共检出1693(23.67%)个存在差异表达的lncRNAs。基因芯片分析结果显示,在表达谱方面,RA组与对照组差异有统计学意义(P<0.05)。与对照组比较,RA组1693个lncRNAs表现出显著性差异表达,在总lncRNAs中占比为23.67%,其中376个lncRNAs的表达上调超过2倍,1317个lncRNAs表现为表达下调;11个lncRNAs表达上调超过了4倍;35个lncRNAs表达下调超过了7倍。GO富集分析显示,376个高表达基因影响到分子功能284项、生物学过程1455项以及细胞组成253项。KEGG Pathway分析显示,主要与脂肪酸的生物合成、胰岛素抵抗通路、腺苷酸活化蛋白激酶信号通路、内吞作用通路等有关。结论 RA患者血清中差异表达的lncRNAs对RA的发生和发展可能起到一定的作用。 Objective Serum long-chain non-coding RNA is closely related to the development of rheumatoid arthritis(RA). This article aims to investigate the differential expression of lncRNA in the serum of patients with rheumatoid arthritis. Methods We collected the serum samples of RA patients and outpatient health examiners admitted to the General Hospital of the Eastern Theater Command from November 2017 to November 2018. Among them, 30 serum samples of RA patients(RA group), 30 serum samples of healthy persons(control group). The differential expression and bioinformatics analysis of LncRNAs in the serum of 30 RA patients and 30 health check-up personnel were carried out by using gene chip technology(CapitalBio Technology Human LncRNA Array v4, 4 x180 K chip). Results A total of 1693(23.67%) differentially expressed lncRNAs were identified, accounting for 23.67% of all lncRNAs. The results of the gene chip analysis showed that in terms of the expression profile, the difference between the RA group and the control group was statistically significant. Among them, 376 lncRNAs were highly expressed in RA serum(fold change> 2, P<0.05), and 1317 lncRNAs were poorly expressed(fold change> 2, P<0.05);There were 11 lncRNAs with over 4-fold changes in high expression and 35 lncRNAs with over 7-fold changes in low expression. The GO and KEGG pathway analysis of lncRNAs with statistically significant differences in expression profiles showed that 376 up-regulated genes participated in 1455 biological processes, 284 molecular functions, and 253 cell compositions. KEGG pathway analysis may be mainly related to fatty acid biosynthesis, insulin resistance pathway, adenylate activated protein kinase signal pathway, endocytosis pathway, etc. Conclusion The differentially expressed lncRNAs in the serum of RA patients may play a certain role in the occurrence and development of RA.
