淫羊藿苷对大鼠庆大霉素耳毒性的防护作用 被引量:2

摘要 目的:研究淫羊藿苷(ICA)对大鼠庆大霉素(GM)耳毒性的防护作用。方法:SPF级雌性SD大鼠,随机分为对照组、模型组、淫羊藿苷组;对照组给予生理盐水[10 mL/(kg·d)]灌胃;模型组先给予对照组等量生理盐水灌胃,1 h后腹腔注射GM注射液[100 mg/(kg·d)];淫羊藿苷组先给予ICA[60 mg/(kg·d)]灌胃,1 h后腹腔注射与模型组等量GM注射液,1次/d,连续14 d。造模结束后,各组给予相应药物治疗28 d。HE染色观察大鼠耳蜗组织病理改变;ELISA检测大鼠血清超氧化物歧化酶(SOD)活性及一氧化氮(NO)、25-羟维生素D3[25(OH)D3]、骨钙素(BGP)、雌二醇(E2)、卵泡刺激素(FSH)的水平;免疫组化检测耳蜗组织CYP27B1、CYP24A1蛋白的表达情况。结果:与对照组比较,模型组大鼠HE染色光镜下可见耳蜗组织产生不同程度的病理损伤,血清SOD活性及25(OH)D3、BGP、E2水平均明显降低,NO、FSH水平明显升高(P<0.05或P<0.01);耳蜗组织CYP27B1蛋白表达降低,CYP24A1蛋白表达升高(P<0.05或P<0.01)。与模型组比较,淫羊藿苷组HE染色光镜下可见耳蜗组织病理损伤较模型组轻,血清SOD活性和25(OH)D3、BGP、E2的水平均升高,NO、FSH水平降低(P<0.05或P<0.01);耳蜗组织CYP27B1蛋白表达明显升高,CYP24A1蛋白表达明显降低(P<0.01)。结论:ICA可有效防护大鼠GM所致耳毒性损伤,其机制可能与抗氧化损伤,影响维生素D、骨钙素、雌激素水平及调控维生素D相关蛋白表达有关。 Objective: To study the protective effect of icariin(ICA) on the ototoxicity of gentamicin(GM) in rats. Methods: SPF female SD rats were randomly divided into control group, model group, and icariin group.The control group was given normal saline 10 mL/(kg·d) by gavage,the model group was given the same amount of normal saline in the control group, and then the GM injection of 100 mg/(kg·d) was intraperitoneally injected1 h later. The icariin group was given ICA 60 mg/(kg·d) by gavage first, and the same amount of GM injection as the model group was intraperitoneally injected 1 hour later, once a day for 14 consecutive days. After modeling,each group was given corresponding medication for 28 days. He staining was used to observe the pathological changes of cochlea in rats. The activities of superoxide dismutase(SOD) and the levels of nitric oxide(NO),25(OH) D3, osteocalcin(BGP), estradiol(E2) and follicle stimulating hormone(FSH) levels were detected by ELISA. The expression of CYP27 B1 and CYP24 A1 in cochlea was detected by immunohistochemistry. Results:Compared with the control group, the pathological damage of cochlea tissue was observed in the model group under HE staining light microscope. The serum SOD activity and 25(OH) D3, BGP, E2 levels were significantly decreased, and the levels of NO and FSH were significantly increased(P <0.05 or P <0.01). The expression of CYP27 B1 protein was decreased and the expression of CYP24 A1 protein was increased(P <0.05 or P <0.01).Compared with the model group, the pathological damage of cochlea tissue in icariin group was lighter than that in model group, serum SOD activity and 25(OH) D3, BGP, E2 levels were increased, NO and FSH levels were decreased(P<0.05 or P<0.01). The expression of CYP27 B1 protein was significantly increased, and the expression of CYP24 A1 protein was significantly decreased(P<0.01). Conclusion: ICA can effectively protect the ototoxic damage caused by GM in rats, and its mechanism may be related to anti-oxidative damage, affecting the levels
