摘要
目的运用网络药理学的方法探讨青风藤治疗类风湿关节炎的分子机制。方法通过TCMSP平台筛选青风藤活性成分及作用靶点;在GeneCards和OMIM数据库中获取类风湿关节炎靶基因。通过R语言对基因进行匹配运算并绘制韦恩图;在String平台生成成分-蛋白互作网络图;Cytoscape 3.6.1软件构建成分-靶点网络;通过R3.5.1软件进行GO富集分析和KEGG通路富集分析。结果共筛选出6个活性成分和23个潜在作用靶点;GO功能富集显示,青风藤可以影响炎症因子的细胞信号转导等生物学过程。KEGG通路富集分析显示,青风藤可以通过调控P53等凋亡相关通路发挥抗类风湿关节炎的作用。结论青风藤可能通过作用于CASP3、JUN、OPRD1等关键靶点调控P53等信号通路,从而调控炎症细胞凋亡进而发挥治疗类风湿关节炎的作用。
Objective To explore the molecular mechanism of Qingfengteng in the treatment of rheumatoid arthritis using the network pharmacology method.Methods The active ingredients and targets of Qingfengteng were screened through the TCMSP platform;target genes of rheumatoid arthritis was obtained from GeneCards and OMIM databases.The gene was matched by R language and a Venn diagram was drawn;the component-protein interaction network diagram was generated on the String platform;the component-target network was constructed by Cytoscape 3.6.1 software;GO enrichment analysis and KEGG pathway enrichment analysis were carried out by R3.5.1 software.Results A total of 6 active ingredients and 23 potential targets were screened;GO function enrichment showed that Qingfengteng can affect biological processes such as cell signal transduction of inflammatory factors.Enrichment analysis of KEGG pathway showed that Qingfengteng can play an anti-rheumatoid arthritis role by regulating apoptosis related pathways such as P53.Conclusion Qingfengteng may regulate P53 and other signaling pathways by acting on key targets such as CASP3,JUN,and OPRD1 and so on,thereby regulating inflammatory cell apoptosis and playing a role in the treatment of rheumatoid arthritis.
