摘要
目的探讨穿心莲内酯(AG)联合氨苄青霉素(AMP)对奈瑟球菌感染脑膜炎大鼠的治疗作用及其机制。方法将100只SD大鼠随机分为正常对照组、模型组、AG组(50mg/kg)、AMP组(200mg/kg)和联合组(AG 50mg/kg+AMP 200mg/kg)共5组,每组各20只。除正常对照组外,其余各组大鼠通过小延髓池注射奈瑟球菌悬液复制脑膜炎大鼠模型。造模后,各组均1次/12小时给药,共给药5次。统计各组大鼠存活率,行症状评分,检测脑组织含水量,ELISA法检测血浆脂多糖和脑组织TNF-α、IL-1β、IL-6含量,Western blot法检测脑组织p65、IκBα蛋白表达。结果与模型组比较,AG组、AMP组、联合组大鼠存活率显著升高(P<0.01),症状评分、脑组织含水量显著降低(P<0.05),血浆脂多糖和脑组织TNF-α、IL-1β、IL-6含量显著降低(P<0.01),p65表达显著下调而IκBα表达显著上调(P<0.01)。与AG组和AMP组比较,联合组大鼠存活率显著升高(P<0.01),症状评分和脑组织含水量显著降低(P<0.05)。与AMP组比较,AG组和联合组血浆脂多糖和脑组织TNF-α、IL-6含量显著降低(P<0.05),p65表达显著下调而IκBα表达显著上调(P<0.05)。结论AG联合AMP能够降低奈瑟球菌感染脑膜炎大鼠病死率,改善神经症状,抑制脑组织水肿。其机制可能与抑制内毒素血症和NF-κB炎性通路,进而抑制炎性反应有关。
Objective To investigate the therapeutic effect and mechanism of Andrographolide(AG)combined with Ampicillin(AMP)on neisseria meningitidis rats.Methods 100 SD rats were randomly divided into normal control group,model group,AG group(50 mg/kg),AMP group(200 mg/kg)and combined group(AG 50 mg/kg+AMP 200 mg/kg),n=20.Except the normal control group,the rats in other groups were injected neisseria suspension into the medulla oblongata to establish the meningitis rat models.After modeling,the drugs were given once per 12 hours for five times.The survival rate of the rats in each group was calculated;the symptom score and the water content of brain tissue were measured;the contents of lipopolysaccharide in plasma and TNF-α,IL-1β,IL-6 in brain tissue were detected by ELISA method.The expression of P65,IκBαwere detected by Western blot method.Results Compared with the model group,the survival rate in AG group,AMP group and combined group were significantly increased(P<0.01),and the symptom score and the water content of brain tissue were significantly decreased(P<0.05).The contents of lipopolysaccharide in plasma and TNF-α,IL-1β,IL-6 in brain tissue were significantly decreased(P<0.01).The expression of P65 was significantly down-regulated while the IκBαwas up-regulated(P<0.01).Compared with the AG group and AMP group,the survival rate in combined group was significantly increased(P<0.01),and the symptom score and water content were significantly decreased(P<0.05).Compared with the AMP group,the contents of lipopolysaccharide in plasma and TNF-α,IL-6 in brain tissue of AG group and combined group were significantly decreased(P<0.05),and the expression of P65 was significantly down-regulated while the IκBαwas significantly up-regulated(P<0.05).Conclusion AG combined with AMP can reduce the mortality rate of neisseria meningitidis rats,improve neurological symptoms and inhibit brain tissue edema.The mechanism may be related to the inhibition of endotoxemia and NF-κB inflammation pathway,and thus inhibiting inflam
