梅山猪与杜洛克猪卵泡中circ0010513的鉴定及分析 被引量:2

Identification and analysis of circ0010513 in follicles of Meishan and Duroc pigs

摘要 试验旨在探索circRNA在卵泡中可能发挥的潜在作用,为了解circRNA调控卵泡发育的机制奠定基础。采用RNA-Seq和生物信息学方法鉴定梅山猪与杜洛克猪卵泡中circRNA的表达量,运用miRanda、RNAhybrid和targetscan软件,对差异性表达的circRNA进行靶基因预测;对靶基因进行GO和KEGG富集分析,并运用qRT-PCR分析梅山猪和杜洛克猪circ0010513在各时期卵泡及各组织的表达量。结果表明:circ0010513在梅山猪M2卵泡(M2卵泡直径5.1~7.0 mm)中的表达量极显著高于杜洛克猪(P<0.01),而梅山猪M1(M1卵泡直径3.1~5.0 mm)卵泡中的表达量极显著高于杜洛克猪(P<0.01),该结果与高通量测序结果一致。circ0010513在心脏、肝脏、脾脏、肺脏、肾脏、下丘脑、垂体、卵巢、输卵管、子宫和子宫角中均有不同程度的表达,其潜在靶基因DUSP5、GADD45G、LAMB3等通过参与繁殖相关的信号通路影响猪的卵泡发育,提示其可能通过对靶基因的调控作用间接参与卵泡发育过程。 This study was to explore the potential role of circRNA in follicles and to lay the foundation for understanding the mechanism by which circRNA regulates follicular development.RNA-Seq and bioinformatics methods were used to identify the expression of circRNA in follicles of Meishan and Duroc pigs.Target gene prediction was performed on differentially expressed circRNA using miRanda,RNAhybrid and targetscan,and GO and KEGG enrichment wwas performed on the target genes.The expression levels of circ0010513 in the follicles and tissues of Meishan and Duroc pigs were analyzed by qRT-PCR.The results showed that the expression of circ0010513 in M2 follicles(M2 Follicle diameter:5.1-7.0 mm)of Meishan pigs was significantly higher than that of Duroc pigs(P<0.01),while the expression in M1 follicles(M1 Follicle diameter:3.1-5.0 mm)of Meishan pigs was significantly higher than that of Duroc pigs(P<0.01).Consistent with high-throughput sequencing results,circ0010513 was expressed at different degrees in heart,liver,spleen,lung,kidney,hypothalamus,pituitary,ovary,fallopian tube,uterus and uterine horn.Its potential target genes,such as dusp5,gadd45g and LAMB3,affect the follicular development of pigs by participating in reproductive related signaling pathways,suggesting that it may be involved in the follicular development process indirectly through regulation of target genes.
