摘要
目的观察茯苓多糖对甲醛诱导DNA-蛋白交联物(DPC)和8-羟基脱氧鸟苷(8-OHDG)的抑制作用,探索茯苓多糖的抗遗传毒性效应。方法将40只成年昆明种雄性小鼠随机分成阳性对照组(甲醛染毒)和茯苓多糖低、中、高剂量组,每组10只。阳性对照组灌胃0.4 ml浓度为0.5 g/L的甲醛溶液,茯苓多糖组小鼠除了灌胃0.2 ml浓度为1.0 g/L的甲醛溶液外,还灌胃0.2 ml不同浓度的茯苓多糖溶液(低剂量组2.0 g/L,中剂量组4.0 g/L和高剂量组8.0 g/L)。1周后摘眼球取血,用ELISA法测定各组小鼠血清DPC和8-OHDG含量。结果各茯苓多糖组DPC和8-OHDG浓度均低于甲醛组;茯苓多糖剂量越高,DPC和8-OHDG浓度越低,呈现明显的剂量-效应关系(DPC:rs=-0.855,P<0.01;8-OHDG:rs=-0.412,P<0.05);DPC与8-OHDG呈正相关关系(r=0.361,P<0.05)。结论茯苓多糖可抑制甲醛诱导小鼠体内DPC和8-OHDG水平,且存在剂量-效应关系,作用随茯苓多糖剂量的提高而增强,表明茯苓多糖具有较强的抗遗传毒性作用。
Objective To observe the inhibitory effect of pachymaran on DPC and 8-OHDG information induced by formaldehyde, and explore its anti-genotoxicity effect. Methods 40 adult male Kunming mice were randomly divided into four groups, in which the positive control group was perfused with 0.4 ml formaldehyde solution of 0.5 g/L, pachymaran low, medium and high dose groups, were respectively perfused with 0.2 ml pachymaran solution of 2,4 and 8 g/L besides 0.2 ml formaldehyde solution of 1.0 g/L. A week later, the blood of the eyeball was taken out to measure the concentration of mice peripheral blood DPC and 8-OHDG by the ELISA method. Results Pachymaran processing groups DPC and 8-OHDG levels were lower than those of the positive control group. A negative correlation was observed between pachymaran dose and DPC and 8-OHDG level(DPC: r_s=-0.855, P<0.01;8-OHDG: r_s=-0.412, P<0.05). A positive correlation between DPC and 8-OHDG was also observed(r=0.361, P<0.05). Conclusion It can be inferred that pachymaran can stop the formation of mice serum DPC and 8-OHDG induced by formaldehyde, and the dose-effect relationship is obvious. The effect can be enhanced with the increase of the pachymaran dose. The conclusion can be drawn that pachymaran has strong anti-genotoxicity effect.
