摘要
目的:应用网络药理学研究琥珀散治疗子宫内膜异位症的作用靶点、活性成分、信号通路等,从而探讨琥珀散治疗子宫内膜异位症的作用机制。方法:应用中药系统药理学分析平台(TCMSP)数据库及文献检索对琥珀散中10味中药主要化学成分进行筛选。采用Pubchem、HIT、SwissTargetPrediction、SEA数据库,对筛选出的化合物进行靶点预测。用DisGeNET、Drugbank、TTD、GSE6364、GSE7305数据库,对子宫内膜异位症进行靶点预测。将药物靶点与疾病靶点进行交集,筛选得到疾病-药物成分共同靶蛋白。运用STRING平台构建蛋白-蛋白相互作用(PPI)网络模型;用Cytoscape 3.7.2软件分别绘制中药-成分网络、成分-靶点网络,进一步筛选核心网络,进行拓扑分析;利用生物信息注释数据库(DAVID)进行基因本体(GO)功能注释富集分析与京都基因与基因组百科全书(KEGG)通路富集分析;利用Cytoscape软件构建琥珀散"有效成分-药物靶点-疾病靶点-信号通路"拓扑网络,确认可能的关键靶点和主要通路。结果:从琥珀散中筛选出190个化合物成分,与子宫内膜异位症关联最强的11个核心靶点;同时筛选出与琥珀散关键成分相关的主要信号通路7条;在此基础上构建了琥珀散的成分-靶点、成分-子宫内膜异位症靶点及相关信号通路的共同表达网络,发现琥珀散可能通过抗黏附、侵袭、种植等多靶点、多途径发挥对子宫内膜异位症的治疗作用。结论:从多角度探索了琥珀散治疗子宫内膜异位症的作用机制,揭示了琥珀散的药理作用,为其后续的实验研究及临床疗效指标的筛选提供了方向。
Objective: To investigate the target,active ingredient and signaling pathway of amber powder in the treatment of endometriosis by using network pharmacology and to explore the mechanism of amber powder in treating endometriosis. Methods: TCMSP database and literature retrieval were used to screen the main chemical components of 10 Chinese medicines in amber powder. Target prediction of the selected compounds was performed using Pubchem,HIT,SwissTarget Prediction and SEA database. Target prediction of endometriosis was performed using DisGeNET,Drugbank,TTD,GSE6364 and GSE7305 databases. The intersection of drug targets and disease targets was carried out to screen the common target proteins of disease-drug components. The STRING platform was used to construct the protein-protein interaction( PPI) network model. Cytoscape 3.7.2 software was used to draw TCM-component network and component-target network respectively to further screen the core network for topology analysis. The gene ontology( GO) functional annotation enrichment analysis and the Kyoto encyclopedia of genes and genomics( KEGG) pathway enrichment analysis were performed by using the bioinformatics annotation database( DAVID). Cytoscape software was used to construct amber powder’s topological network of active ingredient-drug target-disease target-signal pathway to identify possible key targets and main pathways. Results: Totally 190 compounds were selected out from amber powder,including 11 core targets with the strongest correlation with endometriosis. At the same time,7 signal pathways related to the main components of amber powder were screened out. On this basis,a common expression network of the component-target,component-endometriosis target and related signal pathways of amber powder was constructed,and it was found that amber powder may play a therapeutic role in endometriosis through anti-adhesion,invasion,implantation and the other multi-target and multi-pathway. Conclusion: The mechanism of amber powder in the treatment of endometriosis
