多效唑(PP333)对铁皮石斛瓶内开花的促进作用 被引量:5

Enhancement of Paclobutrazol on Flowering in Vitro of Dendrobium officinale Kimura et Migo

摘要 为了研究不同浓度多效唑PP333和不同预处理时间对铁皮石斛瓶内开花的影响。本研究以无菌铁皮石斛(Dendrobium officinale Kimura et Migo)原球茎为试验材料,在培养基中添加不同浓度的多效唑,并设定不同的预培养时间。结果显示,对照处理(未添加多效唑)的铁皮石斛组培苗未出现开花现象,而在培养基中添加不同浓度的多效唑后,铁皮石斛组培苗开花率均显著提高。当多效唑浓度为1.0 mg/L、预培养时间为30 d时,铁皮石斛组培苗开花率达最大(62.50%),花期最长(59.33 d);当多效唑浓度为1.0 mg/L、预培养时间为50 d时,铁皮石斛组培苗始花期最早(21.67 d),花朵品质最好。因此,本研究获得了石斛瓶内开花的最适PP333处理浓度和最佳预培养时间,为铁皮石斛试管花卉的开发提供参考,同时为其他兰科植物的瓶内开花提供理论和技术指导。 In order to s tudy the effects of different concentrations of paclobutrazol and pretreatment time on flowering in vitro of Dendrobium officinale Kimura et Migo.Using PLBs(protocorm-like-body)of sterile D.officinale Kimura et Migo as the experimental materials,with adding different concentrations of paclobutrazo to the medium and setting different pre-culture times.The results showed that no flowering phenomenon was found in tissue culture plantlets of D.officinale Kimura et Migo with no paclobutrazol in the medium(as the control).However,when different concentrations of paclobutrazol were added in the medium,the flowering rate of tissue culture plantlets of D.officinale Kimura et Migo was significantly increased.When paclobutrazo concentration was 1.0 mg/L and the pre-culture time was 30 d,the flowering rate reached the maximum(62.50%),the plantlets were 59.33 d;when paclobutrazo concentration was 1.0 mg/L and the pre-culture time was 50 d,and the day to first flower bud was 21.67 d and the best quality flower was obtained.Therefore,this study obtained the optimal PP333 treatment concentration and the best pre-culture time on flowering in vitro of D.officinale Kimura et Migo,which provided reference for the development of the test tube flower of D.officinale Kimura et Migo,and provided theoretical and technical guidance for the flowering of other orchids.
