摘要
为深入理解光信号通路关键转录因子基因PIF3在丹参中的生物学作用,本研究通过克隆获得SmPIF3基因的编码序列,并对其序列特征和表达特性进行分析。结果表明,该序列包含1个2085 bp的开放阅读框(ORF),编码695个氨基酸,蛋白定位于细胞核且具有碱性螺旋-环-螺旋(Basic Helix-Loop-Helix)结构域,是典型的bHLH类转录因子。SmPIF3基因在根、茎、叶和花中均有表达,且叶和花中表达量较高。高强度的白光可促进该基因的表达,而高强度的蓝光和红光则抑制其表达。
In order to further understand the biological roles of PIF3 gene,the key transcription factor of light signaling pathway,in Salvia miltiorrhiza,we cloned its homolog gene SmPIF3 and analyzed its sequence as well as expression characteristics in Salvia miltiorrhiza.The results showed that this protein contained a 2085 bp open reading frame(ORF)encoding 695 amino acids.SmPIF3 protein localized in nucleus and contained the basic HelixLoop-Helix structural motif.It was a typical member of the bHLH transcription factor family.SmPIF3 is expressed in roots,stems,leaves and flowers,with higher expressions in leaves and flowers.The expression of the gene was increased by high intensity of white light,while decreased by high intensity of blue light and red light.
