盐酸石蒜碱诱导人食管癌细胞Eca-109凋亡的分子机制 被引量:3

摘要 为探究盐酸石蒜碱诱导人食管癌细胞Eca-109凋亡的分子机制,本实验采用流式细胞术检测不同浓度盐酸石蒜碱对Eca-109细胞凋亡率和细胞周期的影响,并通过Western blot检测凋亡相关信号通路及蛋白的表达。结果显示:盐酸石蒜碱能够将Eca-109细胞阻断在G1期;Bcl-2、Caspase-9表达量降低,p53、c-Caspase-9、p-GSK-3β、p-SAPK/JNK表达量增加,Akt、p-Akt蛋白含量无明显变化。表明盐酸石蒜碱可能通过SAPK/JNK/GSK-3β信号通路抑制食管癌细胞Eca-109细胞周期并诱导其凋亡。 In this experiment,flow cytometry is employed to detect the effect of lycorine hydrochloride of different concentrations on the apoptosis rate and cell cycle of esophageal cancer cell Eca-109,and the signaling pathway of apoptosis and expression of apoptosis-related proteins are detected by western blot to clarify its molecular mechanism of inducing apoptosis of Eca-109 cells.The results show that LH blocks Eca-109 cells in G1 phase;the expression quantity of Bcl-2 and Caspase-9 have decreased while that of p53,c-Caspase-9,p-GSK-3βand p-SAPK/JNK have increased.There was no significant change in the protein content of Akt and p-Akt.The results indicate that LH may inhibit the cell cycle of esophageal cancer cell Eca-109 and induce its apoptosis through SAPK/JNK/GSK-3βsignaling pathway.
