摘要
目的观察石蒜碱衍生物LY-09对人肺癌细胞A549和人乳腺癌细胞MCF-7细胞增殖、迁移的影响,初步探讨其可能的作用机制。方法已上市药物阿霉素和SAHA(组蛋白去乙酰化酶抑制剂)为阳性对照药,对数期稳定生长的A549细胞和MCF-7细胞分别加入不同浓度(9,3,1,0.3,0.1,0.03,0.01,0.003μM)的LY-09进行培养。MTT法检测LY-09对肿瘤细胞增殖的影响,细胞划痕实验检测LY-09对肿瘤细胞迁移能力的影响,考察LY-09分别与阿霉素、SAHA联合使用对肿瘤细胞的影响。结果LY-09可显著抑制A549和MCF-7的增殖(EC 50分别为0.081±0.024和0.275±0.083μM),呈浓度依赖性;LY-09还可以抑制A549和MCF-7的细胞迁移;LY-09与SAHA联合用药可以发挥协同作用。结论石蒜碱衍生物LY-09可以抑制肿瘤细胞的增殖与迁移,有望进一步发展成为抗肿瘤药的化学前体。
Objective To investigate the effects of Lycorine derivative LY-09 on proliferation and migration ofhuman A549 and MCF-7 cells,and preliminarily explore its possible mechanism.Methods With the doxorubicin and SAHA as the positive control,various concentrations(9,3,1,0.3,0.1,0.03,0.01,0.003μM)of LY-09 was added in A549 and MCF-7 cells and then were cultured.MTT assay and the cell wound healing assay were applied to detect the proliferation and migration of cancer cells,respectively.In addition,the combination of LY-09 with doxorubicin or SAHA was also investigated.Results LY-09 could inhibit the proliferation and migration of A549 and MCF-7 cells(EC 50:0.081±0.024;0.275±0.083μM),with the concentration dependent property.The combination of LY-09 and SAHA could play a synergistic roleagainst cancer cells.Conclusion LY-09,a derivative of lycorine,can inhibit the proliferation and migration of A549 and MCF-7 cells and is expected to become a chemical precursor of anti-cancer drugs.
