摘要
目的:采用高效液相色谱法研究甘草酸在人体内对硝苯地平药代动力学的影响。方法:采用Shimadzu C18(250 mm×4.6 mm,5μm)色谱柱,以地西泮为内标,甲醇-水为流动相,流速1.0 mL·min-1,柱温30℃,检测波长238 nm,进样量20.0μL。将16名受试者随机分为实验组和对照组,分别服用甘草酸二铵胶囊(每天3次,每次150 mg)和安慰剂,疗程14d。2组均于第1日晨和第14日晨服药后继续口服硝苯地平控释片30 mg,并在连续服药后1、1.5、2、4、6、8、12、18、24、36、48、60 h同时采集肘部静脉血5 mL,测定硝苯地平的浓度,由Winonlin 6.3计算药代动力学参数。结果:单次给药后Tmax、Cmax、AUC0-60h、AUC0-∞、t1/2、MRT无明显变化(P>0.05);多次给药后实验组硝苯地平的Tmax、Cmax、AUC0-60h、AUC0-∞均明显低于对照组(P<0.05)。结论:连续给予甘草酸可降低人体内硝苯地平的生物利用度。
Objective:To study the effect of glycyrrhizic acid on the pharmacokinetics of nifedipine in vivo by HPLC.Methods:Using Shimadzu C18(250 mm×4.6 mm,5μm)column with diazepam as internal standard and methanol-water as mobile phase,the flow rate was 1.0 mL·min-1,column temperature was 30℃,detection wavelength was 238 nm,and injection volume was 20.0μL.16 subjects were randomly divided into experimental group and control group.They were given diammonium glycyrrhizinate capsules(150 mg each time,3 times a day)and placebo for 14 days respectively.Both groups continued to take 30 mg nifedipine controlled-release tablets on the morning of the first day and the fourteenth day,and at 1,1.5,2,4,6,8,12,18,24,36,48 and 60 h after continuous administration,5 mL of blood in the elbow was collected to determine the concentration of nifedipine.The pharmacokinetic parameters were calculated by Winonlin 6.3.Results:There were no significant changes in Tmax,Cmax,AUC0-24 h,AUC0-∞,t1/2 and MRT after single administration(P>0.05),but the Tmax,Cmax,AUC0-24 hand AUC0-∞of nifedipine in the experimental group were significantly lower than those in the control group(P<0.05).Conclusion:Continuous administration of glycyrrhizic acid could reduce the bioavailability of nifedipine in human body.
