联合应用神经生长因子和血小板衍生生长因子对骨折愈合的影响及机制 被引量:11

Analysis of the effect of nerve growth factor and platelet derived growth factor on fracture healing

摘要 目的探讨神经生长因子(NGF)及血小板衍生生长因子(PDGF)对骨折的愈合影响,以及NGF与PDGF在骨折的愈合中是否有协同效应。方法将48只雄性8周龄SD大鼠(北京斯贝福生物有限公司),采用随机数字表法分为4组,每组12只,分别于骨折断端局部应用NGF(NGF组)、PDGF(PDGF组)、NGF+PDGF(联合组)、生理盐水(对照组)。各组分别于术后14 d和28 d选取6只大鼠,检测血清中骨形态发生蛋白-2(BMP-2)及血管内皮生长因子(VEGF)含量。处死后检测骨痂中碱性磷酸酶(ALP)含量,苏木精-伊红(HE)染色测定骨痂平均骨小梁宽度(TW)和骨小梁表面成骨细胞指数(IOB),组间比较采用方差分析,组间采用SNK法进行两两比较。结果术后14 d,NGF组、PDGF组、联合组、对照组血清中VEGF的含量依次为(235.25±16.33)、(245.40±17.84)、(351.45±17.19)、(212.21±17.96)pg/ml,实验组均较对照组升高,且联合组最高(F=10.459,P<0.05)。术后28 d,NGF组、PDGF组、联合组、对照组骨痂中IOB依次为(356.94±20.30)、(347.35±17.62)、(397.44±22.35)、(301.68±14.20)mm^2,其中联合组高于NGF组、PDGF组和对照组(F=54.992,P<0.05)。结论NGF及PDGF局部应用对骨折愈合均有促进作用,且NGF及PDGF联合应用对骨折愈合具有协同作用,其协同作用机制可能与促进VEGF表达有关。 Objective To investigate the effect of nerve growth factor(NGF)and platelet-derived growth factor(PDGF)on fracture healing,and whether NGF and PDGF have synergistic effect in fracture healing.Methods A total of 48 male 8-week-old SD rats were purchased from Beijing Sibeifu Biological Co.,LTD.,and divided into 4 groups with 12 rats in each group by random number table method.A fracture model was established,and local application of NGF,PDGF,NGF+PDGF and normal saline was performed in groups A,B,C and D,respectively.Six rats in each group were selected 14d and 28 d after surgery,respectively,to detect the contents of bone morphogenetic protein-2(BMP-2)and vascular endothelial growth factor(VEGF)in serum.After death,alkaline phosphatase(ALP)content in callus was detected,and mean trabecular width(TW)and surface osteoblastic index(IOB)of trabecular bone were determined by hematoxylin and eosin(HE)staining.Variance analysis was used for data analysis,and SNK method was used for pair comparison between groups.P<0.05 was statistically significant.Results 14 d after surgery,the serum VEGF levels in NGF group,PDGF group,combined group,and control group were(235.25±16.33),(245.40±17.84),(351.45±17.19)and(212.21±17.96)pg/ml,respectively.The levels in the experimental group were all higher than those in the control group,and the levels in the combined group were the highest(F=10.459,P<0.05).28d after surgery,IOB in NGF group,PDGF group,combined group,and control group was(356.94±20.30),(347.35±17.62),(397.44±22.35)and(301.68±14.20)mm^2 successively,among which,IOB in combination group>NGF group>PDGF group>control group(F=54.992,P<0.05).Conclusion Local application of NGF and PDGF can promote fracture healing,and combined application of NGF and PDGF may have a synergistic effect on fracture healing,and the synergistic mechanism may be related to promoting VEGF release and improving blood supply around the fracture.
