Study on the mechanism of anti-osteoporosis of Semen Sojae Preparatum

摘要 目的:观察豆豉对去卵巢大鼠骨质疏松的影响,初步探讨豆豉治疗骨质疏松症的作用及机制。方法:采用去势法建立骨质疏松症大鼠模型,并灌胃给予豆豉治疗12周,检测大鼠骨密度(BMD)和骨矿物质含量(BMC)、子宫指数和血清中雌二醇(E_2)含量,采用最佳骨组织RNA提取方法,应用RT-qPCR技术检测大鼠骨组织中ERα、ERβmRNA表达水平。结果:与假手术组比较,模型组大鼠BMD和BMC、子宫指数和血清中E_2含量显著降低(P<0.01),ERα、ERβmRNA表达水平也显著降低(P<0.01),给予豆豉治疗后,大鼠BMD和BMC、子宫指数和血清中E_2含量升高,其中,豆豉高剂量组大鼠血清中4个指标较模型组显著升高(P<0.05,P<0.01);各剂量豆豉组ERα、ERβmRNA表达量显著升高(P<0.01,P<0.05),并呈剂量依赖性。结论:豆豉可以调节大鼠体内雌激素分泌水平,提高大鼠骨密度和骨矿物质含量以及血清中E_2含量,改善去卵巢大鼠的骨代谢,对骨质疏松症具有一定的治疗作用。 Objective:To observe the effects of Semen Sojae Preparatum on osteoporosin ovariectomized rats,preliminary investigate the role and mechanism of action of Semen Sojae Preparatumin the treatment of osteoporosis.Methods:The osteoporosis model of ovariectomized rats was established by Castration,and given Semen Sojae Preparatum by intragastric administration,measured the bone mineral density(BMD),bone minerals content(BMC)and uterine index,estradiol(E2)content in serum,using the best bone tissue RNA extraction method,RT-q PCR technique was used to detect the expression of ERαand ERβmRNA in rat bone tissue.Results:Compared with sham group,the BMD,BMC and uterine index,E2 content in serum of model group were significantly decreased(P<0.01).The expression levels of ERαand ERβmRNA were also significantly decreased(P<0.01).After the administration of Semen Sojae Preparatum,the BMD,BMC,uterine index and serum E2 levels in rats were significantly increased.The serum levels in the high dose groups of soybean meal increased significantly(P<0.05,P<0.01).And the expression levels of ERα,ERβmRNA in all doses groups increased significantly(P<0.05,P<0.01),and it was dose-dependent.Conclusion:Semen Sojae Preparatumcan regulate the secretion of estrogen in rats,increase BMD and BMC,and the E2 content in serum,improve bone metabolism in ovariectomized rats,and have certain therapeutic effects on osteoporosis.
