摘要
目的:研究天麻提取物对帕金森病大鼠神经行为学障碍的改善作用。方法:采用两点法于中脑腹侧被盖部和黑质致密部注射6-羟基多巴胺(6-OHDA)复制大鼠帕金森病模型。灌胃给予天麻提取物,每日1次,连续21 d。以盐酸去水吗啡腹腔注射诱导大鼠旋转,观察注射5min后30min内大鼠恒定向健侧旋转的转圈数;以Morris水迷宫实验观察大鼠在平台所在象限的距离百分比(PT%)和停留时间百分比(T%),共同评价天麻提取物对帕金森病大鼠神经行为学障碍的改善作用。结果:与模型对照组比较,天麻提取物低剂量组(5 g生药/kg)、中剂量组(10 g生药/kg)和高剂量组(20 g生药/kg)有降低去水吗啡所诱导的帕金森病大鼠转圈数的趋势(P> 0. 05);天麻提取物高剂量组(20 g生药/kg)在水迷宫的平台象限距离百分比(PT%)和停留时间百分比(T%)明显增加(P <0. 05)。结论:天麻提取物能明显改善帕金森病大鼠的神经行为学障碍,显示出明显的抗帕金森病的作用。
Objective TO study Gastrodia extract on the anti-parkinson s effect.Methods A two-point method was used to inject 6-hydroxydopamine(6-OHDA)into ventral tegmental area(VTA)and substantia nigra pars compacta(SNc)of rats to copy models of parkinson s disease.Gastrodia extract was given by gavage,once per day,continuously for 21 days.The rat rotation was induced by intraperitoneal injection of 0.5mg/kg apomorphine hydrochloride(APO),and the numbers of constant rotation to the healthy side were observed within 30 min after 5 min of injection.Morris water maze experiment was used to observe the percentage of distance(PT%)and the percentage of time(T%)that rats stay in the quadrant where the platform is located.To jointly evaluate the improvement effect of Gastrodia extract on neurobehavioral disorders in parkinson s disease rats.Results Compared with the model group,the low-dose group(5 g crude drug/kg),medium-dose group(10 g crude drug/kg),and high-dose group(20 g crude drug/kg)of Gastrodia extract reduced the number of rotation induced by APO(P>0.05).The high-dose group of Gastrodia extract(20 g crude drug/kg)increased significantly PT%and T%(P<0.05).Conclusion Gastrodia extract can significantly improve the neurobehavioral disorders of parkinson s disease rats,and shows obvious anti-parkinson s effects.
