摘要
目的探讨细胞色素氧化酶 P450-2E1(CYP2E1)在非酒精性脂肪性肝病大鼠中的作用机制,以及茵陈蒿汤对非酒精性脂肪肝病的治疗效果。方法把50只SD大鼠用随机法分成空白对照组15只和实验组35只,空白对照组大鼠饲喂普通饲料,实验组大鼠饲喂改良以后的高脂饲料,并且进行非酒精性脂肪肝病大鼠造模,饲喂24周。待造模成功后,将剩余的实验组大鼠随机分为3组,非酒精性脂肪肝组、辛伐他汀对照组(SVT)和茵陈蒿汤组,每组10只,对大鼠进行灌胃,给予相应的药物治疗,6周后处死大鼠,检测大鼠的低密度脂蛋白胆固醇(LDL-c)含量、肝质量、三酰甘油(TG)、体质量、肝指数、总胆固醇(TC),使用PCR检测肝组织中CYP2E1的表达。结果与空白对照组比较,非酒精性脂肪肝组大鼠的TG、LDL-c、体质量、肝指数、肝质量、TC、CYP2E1的表达均升高(P<0.05);与非酒精性脂肪肝组比较,SVT组、茵陈蒿汤组大鼠各项指标及CYP2E1表达均降低(P<0.05);与SVT组比较,茵陈蒿汤组各项指标及CYP2E1表达均降低(P<0.05)。结论茵陈蒿汤可通过调节大鼠CYP2E1的表达水平,减轻脂质异常代谢,降低组织炎症反应,改善肝功能,保护肝组织。
Objective To investigate the mechanism of P450-2E1(CYP2E1) in non-alcoholic fatty liver disease rats and the effect of Yinchenhao decoction on non-alcoholic fatty liver disease.Methods Fifty SD rats were randomly divided into a control group of 15 and an experimental group of 35.The rats in the control group were fed with ordinary feed,and the rats in the experimental group were fed with modified high-fat diet and non-alcoholic.Rats with fatty liver disease were modeled and fed for 24 weeks.After successful modeling,the remaining experimental rats were randomly divided into 3 groups:non-alcoholic fatty liver group,simvastatin control group (SVT) and Yinchenhao decoction group,10 rats in each group.The rats were sacrificed by intragastric administration,and the corresponding drugs were given.After 6 weeks,the rats were sacrificed,and the rats’ LDL-c content,liver weight,triglyceride (TG),body weight,liver index,total cholesterol (TC),and the expression of CYP2E1 in liver tissue was detected by PCR.Results Compared with the control group,the expressions of TG,LDL-c,body weight,liver index,liver weight,TC,and CYP2E1 in the non-alcoholic fatty liver group were increased (P<0.05).Compared with the SVT group,the rats in the SVT group and the Yinchenhao decoction group had decreased expressions of all indicators and CYP2E1 (P<0.05);compared with the SVT group,the indicators in the Yinchenhao decoction group and expressions of CYP2E1 decreased (P<0.05).Conclusion Yinchenhao decoction can reduce the abnormal metabolism of lipids,reduce tissue inflammation,improve liver function,protect liver tissue and treat nonalcoholic fatty liver disease by regulating the expression level of CYP2E1 in rats.
