摘要
目的:探讨背根神经节(dorsal root ganglion,DRG)GABA转运蛋白1(GABA transporter 1,GAT-1)在神经病理性疼痛发生发展中的作用。方法:雌性SD大鼠60只,随机分为4组:对照组(Sham)﹑CCI组﹑CCI+NS组和CCI+NO-711组,每组15只。CCI组和Sham组术后第3、7、14天取DRG,行免疫荧光和Western blot检测GAT-1;CCI+NS组和CCI+NO-711组在CCI手术当天和术后第7天,分别L5 DRG局部注射生理盐水和等体积的GAT-1抑制剂NO-711,测出给药前后机械缩足反射阈值(mechanical withdrawal threshold,MWT)和热缩足反射潜伏期(thermal withdrawal latency,TWL)及DRG内p-ERK、脊髓背角c-Fos的表达。结果:与Sham组相比,CCI组大鼠术后第7天及14天GAT-1升高;相比CCI+NS组,CCI+NO-711组两个时间点DRG给药后TWL和MWT均升高,背根神经节p-ERK及脊髓背角c-Fos的表达降低。结论:神经损伤后DRG中GAT-1表达升高,在神经病理性疼痛发生发展中起着重要作用。
Objective:To investigate the role of GABA transporter 1(GAT-1)in the dorsal root ganglion(DRG)in the pathogenesis of neuropathic pain.Methods:Sixty female Spray-Dawley rats were randomly divided into four groups(n=15 each):Control group(Sham),CCI group,CCI+NS group,CCI+NO-711 group.On the 3 d,7 d,14 d after operation,rats were sacrificed.Western blot and immunofluorescence were used to measure the expression of GAT-1 protein in L4-L5 DRG of Sham group and CCI group.On the day of CCI operation and 7 d after operation,normal saline was injected into the L5 DRG of CCI+NS group,while isometric NO-711 was locally injected into the L5 DRG of CCI+NO-711 group.The changes of mechanical withdrawal threshold(MWT)and thermal withdrawal latency(TWL),and the expression of p-ERK in DRG and of c-Fos in the spinal dorsal horn of CCI rats were recorded before and after injection.Results:Compared with the Sham group,the expression of GAT-1 protein in DRG was significantly increased in CCI group 7 d,14 d after operation.After local L5 DRG injection of NO-711 at two time points,the TWL and MWT were raised and the expression of p-ERK in DRG and of c-Fos in the spinal dorsal horn were decreased,compared with CCI+NS group.Conclusion:The overexpression of GAT-1 in DRG plays an important role in the development and maintenance of neuropathic pain.
