胄叶线蕨提取物对过氧化氢诱导的人脐静脉血管内皮细胞损伤的保护作用研究 被引量:2

摘要 目的探讨胄叶线蕨提取物对过氧化氢(H2O2)诱导的人脐静脉血管内皮细胞(EVC-304)损伤的保护作用。方法建立H2O2诱导的EVC-304氧化损伤模型进行药物试验,分为空白对照组、模型对照组(H2O2浓度为0.1 mmol/L)、胄叶线蕨提取物组(胄叶线蕨提取物浓度分别为20、40、80 mg/L)。以四甲基偶氮唑蓝(MTT)比色法测定细胞存活率,ELISA法测定试剂盒测定一氧化氮(NO)、乳酸脱氢酶(LDH)含量和谷胱甘肽-S转移酶(GST)、超氧化物歧化酶(SOD)的表达水平,以及丙二醛(MDA)的含量、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)的释放量及活性。结果不同剂量胄叶线蕨提取物组EVC-304细胞OD值、存活率与空白对照组比较,差异无统计学意义(P>0.05)。不同剂量胄叶线蕨提取物组OD值高于模型对照组,抑制率低于对照组(P<0.05)。用胄叶线蕨提取物孵育会明显提高NO含量以及SOD、GST活性,降低LDH表达来增强细胞的抗氧化能力;胄叶线蕨提取物能增强CAT、GSH-Px的活性,降低MDA水平,加速氧自由基清除。结论胄叶线蕨提取物对H2O2诱导的EVC-304氧化应激损伤具有一定保护作用,其机制可能通过抑制细胞内氧化应激水平而起到抗氧化损伤的作用。 Objective To explore the protective effect of Colysis hemitoma extract on oxidative stress injury of human umbilical vein endothelial cells.Methods A H2O2-induced human umbilical vein endothelial cells(EVC-304)oxidative damage model was established for drug testing,it was divided into normal control group,H2O2 damage model group(the concentration of H2O2 was 0.1 mmol/L),and Colysis hemitoma extract group(the mass concentration of total flavonoids from Colysis hemitoma were 20,40,80 mg/L).The cell survival rate was measured by tetramethyl azozo blue(MTT)colorimetric method,and the ELISA method was used to determine the content of nitric oxide(NO),lactate dehydrogenase(LDH)and glutathione in the cell supernatant.-S-transferase(GST),superoxide dismutase(SOD)expression levels,and the content of malondialdehyde(MDA)in cells and catalase(CAT),glutathione peroxidase(GSH-Px)release and activity changes.Results Compared with the blank control group,the OD value and survival rate of EVC-304 cells in different doses of Colysis hemitoma extract group had no significant difference(P>0.05).The OD value of different doses of Colysis hemitoma extract group was higher than model control group,and the inhibition rate was lower than control group(P<0.05).Incubation with Colysis hemitoma extract can obviously increase NO content,SOD and GST activities,and reduce LDH expression to enhance the antioxidant capacity of cells.The Colysis hemitoma extract can enhance the activities of CAT and GSH-Px,reduce the level of MDA and accelerate the scavenging of oxygen free radicals.Conclusion The extract of Colysis hemitoma has a certain protective effect on oxidative stress injury induced by H2O2 in EVC-304,and its mechanism may play a role in preventing oxidative damage by inhibiting the level of oxidative stress in cells.
