血栓闭塞性脉管炎的SNP靶点分析研究 被引量:1

摘要 目的研究血栓闭塞性脉管炎患者的SNP靶点。方法以2016年11月-2019年7月在新疆维吾尔自治区人民医院血管外科和新疆医科大学第一附属医院血管外科就诊的血栓闭塞性脉管炎患者177例为研究对象,以同时期在体检中心86例健康成年男性为对照。采用芯片法筛选差异位点,采用Logistic回归分析,通过质量控制分析确定SNP靶点的研究。对易感SNP所在基因及上下游相关基因进行富集分析,通过富集差异基因的Pathway条目,找寻引起疾病发病及进展的细胞通路。结果通过检验计算SNP与疾病关联的显著性筛选出26个血栓闭塞性脉管炎相关SNP靶点,通过GO与Pathway富集分析,最终获得16个靶点。筛选出26个SNP所在的基因及附近所涉及的基因共84个,其主要作用于细胞粘附,生物粘附,细胞与细胞间粘附,细胞真菌起源的分子应答,调节细胞组件大小,细胞内的信号级联,肿瘤坏死因子生成的正向调控,模式识别受体信号通路,激活先天性免疫,先天性免疫激活反应等生物学过程。结论血栓闭塞性脉管炎与SPN靶点具有相关性。 Objective To investigate SNP targets in the patients with thromboangiitis obliterans(TAO).Methods A total of 177 patients with TAO who visited the Department of Vascular Surgery of Xinjiang Uygur Autonomous Region People’s Hospital and the First Affiliated Hospital of Xinjiang Medical University from November 2016 to July 2019 were studied, and 86 healthy adult males who visited the physical examination center during the same period were selected as control subjects. The SNP screening was used to screen and determine the difference sites, Logistic regression analysis was used to determine the SNP target correlation study through quality control analysis. Enrichment analysis was conducted on the genes of susceptible SNP and the related upstream and downstream genes to find pathway entries for enrichment of the differential genes, and to look for possible functions and cell pathways of the differential genes. Results Twenty-six SNP targets related to TAO were screened by testing and calculating the significance of association between the SNPs and disease. Sixteen targets were finally obtained by GO and pathway enrichment analysis. A total of 84 SNPs were screened out, which mainly involved in cell adhesion, biological adhesion, cell to cell adhesion, molecular response of cell fungal origin, regulation of cell assembly size, intracellular signal cascade, positive regulation of tumor necrosis factor production, pattern recognition receptor signaling pathway, activation of innate immunity and innate immunity activation reaction and other biological processes.Conclusion There is a correlation between TAO and SPN.
