308nm单频准分子光对人角质形成细胞活力及细胞因子分泌水平的影响

摘要 目的探索308nm单频准分子光照射HaCaT细胞后对其细胞活力以及分泌SCF、bFGF、GM-CSF等细胞因子的影响。方法MTT法检测308nm单频准分子光0-600mJ/cm^2的不同照射剂量照射后6h、12h、24h、48h对角质形成细胞的细胞活力的影响;ELISA法检测细胞培养上清液中SCF、bFGF、GM-CSF的浓度。结果0-500mJ/cm^2范围内308nm单频准分子光照射对角质形成细胞活力无明显影响,但600mJ/cm^2照射剂量下细胞活力下降(P<0.05)。308nm单频准分子光照射组的SCF、bFGF、GM-CSF分泌水平较对照组高(P<0.05),在0-400mJ/cm^2照射剂量范围内,SCF、bFGF的分泌水平随着照射剂量的增加而升高(P<0.05),照射12-24h后达高峰水平。各照射剂量组GM-CSF分泌水平无统计学差异(P>0.05)。结论308nm单频准分子光可能通过诱导角质形成细胞分泌SCF、bFGF、GM-CSF等与黑素细胞增殖分化、黑素合成相关的细胞因子,而发挥治疗作用。 Objective To investigate the effects of 308nm monochromatic excimer light(MEL)on the cell viability and secretion of cytokines such as SCF、bFGF、GM-CSF in human keratinocytes.Methods HaCat cells were irradiated at doses of 0mJ/cm^2,100mJ/cm^2,200mJ/cm^2,300mJ/cm^2,400mJ/cm^2,500mJ/cm^2,600mJ/cm^2 using a 308nm MEL and then cultured for an additional 6,12,24,48 hours.Cell viability were measured by MTT methods.The secretion of SCF、bFGF、GM-CSF was analyzes by ELISA.Results Cell viability of keratocytes was not decreased by the exposure to 0-500 mJ/cm^2 of 308nm MEL.The secretion levels of SCF、bFGF、GM-CSF in the 308nm MEL group were significantly higher than those in control group.The secretion levels of SCF、bFGF increased in a dose-dependent manner between 0-400 mJ/cm^2,while The secretion levels of GM-CSF showed no signicant difference between different doses.The secretion levels peaks after 12-24 hours of irradiation.Conclusion 308nm monochromatic excimer light may promote the secretion of SCF、bFGF、GM-CSF of keratocyte,which are associatied with the proliferation and differentiation of MC,the synthesis of melanin.
