17β-羟基类固醇脱氢酶3基因插入/缺失与公猪睾丸表型性状关联研究 被引量:1

Genetic Correlation between Indel Locus of Hsd17b3 Gene and Reproductive Traits in Boar Testis

摘要 17β-羟基类固醇脱氢酶3(Hsd17b3)基因主要表达于哺乳动物睾丸间质细胞(LCs)中,是合成睾酮和维持雄性生殖功能的关键基因之一。为进一步推进猪产业的分子标记辅助育种,该研究探究Hsd17b3基因遗传变异位点与公猪睾丸表型性状的相关性,以期丰富该基因多态位点与公猪繁殖的遗传关联。利用PCR技术在大白猪(n=257)和长白猪(n=62)中检测到Hsd17b3基因第2内含子和第4内含子的两个indel位点,通过琼脂糖凝胶电泳及测序技术验证了该基因L3-10-bp和L11-11-bp的indel突变。关联分析结果表明:L3-10-bp的indel位点与40日龄长白猪的睾丸长轴长、睾丸短轴长和睾丸重均呈显著相关(P<0.01)。该结果与雄性睾丸的早期发育阶段和初始快速发育阶段睾丸发育过程相吻合,提示该indel位点可作为猪标记辅助选择的潜在DNA标记,为Hsd17b3基因在猪遗传育种中的研究和应用提供指导。 17-hydroxysteroid dehydrogenase 3(Hsd17b3)gene,which is mainly expressed in mammalian testicular stromal cells(LCs),is one of the key genes for the synthesis of testosterone and the maintenance of male reproductive function.To explore Hsd17b3 gene's insertion/deletion(indel)loci and boars testicular phenotype correlation,this study using PCR amplification techniques in landrace(n=257)and large white(n=62)detected two indel site in Hsd17b3 gene,verified by agarose gel electrophoresis and sequencing the genes L3-10-bp and L11-11-bp indel mutation.The results of correlation analysis showed that the Indel site of L3-10-bp was significantly correlated with the long axis length,short axis length and testicular weight of 40-day-old long white pigs(P<0.01).The above results were consistent with the early development stage and initial rapid development stage of male testicle,suggesting that the Indel locus could be used as a potential DNA marker for marker-aided selection in pigs.The above findings could provide a certain basis and guidance for the research and application of Hsd17b3 gene in pig genetics and breeding.
