摘要
为了研究儋州鸡与文昌鸡、黎凤鸡、昌江霸王山鸡和北京油鸡等不同地方鸡种群体的遗传多样性及遗传距离,促进海南地方鸡儋州鸡的遗传资源保护和开发利用,试验利用STR分型技术选取15个微卫星位点,对5个群体共240只个体进行遗传多样性检测。结果表明:儋州鸡的平均有效等位基因数(Ne)为2.76个,期望杂合度(He)为0.644 6,多态信息含量(PIC)为0.584 7,其他4个种群中,文昌鸡的Ne、He和PIC最高(3.00、0.650 5和0.597 5),北京油鸡最低(2.66、0.588 2和0.524 3);遗传距离和系统聚类结果显示,5个鸡群体聚为3类,儋州鸡和昌江霸王山鸡2个群体为一类,文昌鸡和黎凤鸡为一类,北京油鸡为一类。说明儋州鸡与霸王山鸡种间遗传关系较近,与其他品种遗传关系较远。
An attempt was made to analyze the genetic diversity and genetic distance of populations of Danzhou chicken,Wenchang chicken,Lifeng chicken,Changjiang Bawang chicken and Beijing-youji chicken to facilitate the protection and utilization of genetic diversity of Danzhou chicken,eA short tandem repeat(ST)typing method was used to select 15 microsatellite loci to detect the genetic diversity of 240 individuals in the 5 chicken populations.Danzhou chicken was found to have an average effective number of alleles(Ne)of 2.76,expected heterozygosity(He)of 0.6446,and polymorphic information content(PIC)of 0.5847.Among the other 4 chicken populations,Wenchang chicken has the highest Ne,He and PIC(3.00,0.6505 and 0.5975,respectively);Beijing-youji chicken has the lowest(2.66,0.5882 and 0.5243,respectively).The results of genetic distance and cluster analysis showed that five chicken populations are grouped into three groups:Danzhou chicken and Bawang chicken in one group,Wenchang Chicken and Lifeng chicken in one group,Beijing-youji chicken in one group.The results showed that Danzhou chicken has a close genetic relationship with Bawangshan chicken is but a far genetic relationship with the other breeds.
