水产源产细菌素乳酸菌的分离、筛选及鉴定 被引量:6

摘要 为拓展水产源产细菌素乳酸菌的开发利用,本研究从水产动物及其生境中分离出乳酸菌261株,以细菌素敏感菌为指示菌,筛选产细菌素菌株,鉴定产生菌,并分析所产细菌素的稳定性。结果表明:(1)5株菌上清液排除有机酸后仍对指示菌有抑制作用,与对照组(pH 6.0)相比,菌株G7-1发酵上清经排酸处理后,抑菌率仅降低了1.59%(P>0.05)。进一步经过氧化氢酶和胃蛋白酶处理后,抑菌率分别显著降低了5.07%和21.54%(P<0.05),表明G7-1发酵上清液的抑菌物质含细菌素或其类似物。(2)经16S rDNA序列同源性分析,鉴定菌株G7-1为屎肠球菌。进一步检测其细菌素的稳定性发现,G7-1所产细菌素对热不稳定,但对酸碱处理有较好的耐受性。 In order to improve the development and application of bacteriocin-producing Lactic acid bacteria from aquatic sources,the present study isolated 261 bacteriocin-producing Lactic acid bacteria strains from aquatic animals and their habitats.Using a bacteriocin-sensitive bacteria as theindicator bacteria,bacteriocin-producing strains were screened,identified,and the stability of its bacteriocin was analyzed.The results showed that:(1)The supernatant of 5 out of the isolated strains had the inhibitory effect against the bacteriocin-sensitive indicator bacteria after neutralizing the organic acid.Compared with the control group(pH 6.0),the bacteriostatic effect of fermentation supernatant from G7-1 was reduced only 1.59%after neutralization(P>0.05).Further treatment with catalase and pepsin of G7-1 supernatant significantly(P<0.05)reduced the inhibition rate by 5.07%and 21.54%,respectively;(2)According to the 16S rDNA sequence homology analysis,G7-1 was identified as Enterococcus faecium.The results of acid-based tolerance and thermal stability of bacteriocin revealed that the bacteriocin produced by G7-1 was unstable to heat,but had good resistance to acid or alkali treatment.
