3-溴丙酮酸通过糖酵解抑制人肺腺癌H1975细胞增殖及诱导凋亡研究 被引量:5

摘要 目的探究3-溴丙酮酸(3-bromopyruvate,3-BP)对肺腺癌H1975细胞抑制增殖和诱导凋亡的影响。方法采用3-BP(0、10、20、40、80、160μmol·L^-1)处理H1975细胞,使用MTT检测3-BP对肺腺癌H1975细胞增殖的影响。采用集落克隆实验观察低浓度的3-BP(0、5、10、20μmol·L-1)对H1975细胞的增殖抑制作用。划痕和Transwell小室实验检测不同浓度3-BP(0、25、50、75μmol·L^-1)3-BP对H1975细胞迁移能力的影响。Annexin V-FITC/PI双染法检测3-BP对肺腺癌H1975细胞凋亡的影响。试剂盒检测3-BP对H1975细胞内活性氧及ATP水平的改变。Western blot检测3-BP对H1975细胞糖酵解及凋亡相关蛋白的影响。结果3-BP对肺腺癌H1975细胞具有明显的增殖抑制作用并明显降低了H1975细胞的迁移能力。3-BP可以明显增加肺腺癌H1975细胞的凋亡并在此过程中伴随着ATP水平的下降和活性氧水平的增高。Western blot检测表明3-BP上调了促凋亡蛋白Bax的表达,抑制了抑凋亡蛋白Bcl-2、糖酵解相关蛋白MCT1、HK-Ⅱ的表达。结论3-BP可以通过抑制糖酵解抑制人肺腺癌H1975细胞增殖并诱导肺腺癌细胞发生凋亡。 Aim To investigate the effect of 3-bromopyruvate(3-BP)on the proliferation inhibition and apoptosis induction of lung adenocarcinoma cell line H1975.Methods 3-BP(0,10,20,40,80,160μmol·L^-1)was used to treat lung adenocarcinoma cell line H1975.MTT was used to detect the effect of 3-BP on the proliferation of lung adenocarcinoma cell line H1975.The inhibitory effect of low concentration 3-BP(0,5,10,20μmol·L^-1)on the proliferation of H1975 cells was observed by colony experiments.The effect of different concentrations of 3-BP(0,25,50,75μmol·L-1)on the migration of H1975 cells was detected by scratch and Transwell cell experiments.The effect of 3-BP on the apoptosis of H1975 cells was detected by Annexin V-FITC/PI double staining.The changes of reactive oxygen species(ROS)and ATP levels in H1975 cells treated with different concentrations of 3-BP were detected by kit.The effect of 3-BP on glycolysis and apoptosis related proteins in lung adenocarcinoma cell line H1975 was detected by Western blot.Results 3-BP could inhibit the proliferation of H1975 cells and reduce its migration ability.3-BP significantly increased the apoptosis of lung adenocarcinoma cell line H1975,accompanied by the decrease of ATP level and the increase of ROS level.Western blot showed that 3-BP increased the expression of Bax and inhibited the expression of Bcl-2,MCT1,HK-Ⅱ.Conclusions 3-BP can inhibit the proliferation and induce the apoptosis of human lung adenocarcinoma cell line H1975 by inhibiting glycolysis.
