虱螨脲原药对SD大鼠两代繁殖毒性试验 被引量:1

Two-generation reproductive toxicity of lufenuron in SD rats

摘要 目的通过两代繁殖毒性试验,获得虱螨脲原药对雌雄性大鼠繁殖能力、子代生长发育毒性信息。方法无特定病原体级健康SD大鼠随机分为对照组和低、中、高3个剂量组,每组60只,雌、雄各半。低、中、高3个剂量组虱螨脲的染毒剂量分别为5.0、20.0和80.0 mg/(kg体质量·d),对照组饲以标准饲料,采用经口喂养的染毒方式,连续喂养8周后交配繁殖。测定亲代和子一代成年大鼠繁殖指数及脑和生殖器官脏器系数等指标,并作常规病理检查;测定子一代和子二代仔鼠的出生指标和生长指标。结果(1)亲代。亲代中剂量组雌鼠体质量从染毒第14天开始均低于对照组雌鼠(P<0.05),从染毒第35天开始均低于低剂量组雌鼠(P<0.05);高剂量组雌鼠体质量从染毒第14天开始均低于其余3组雌鼠(P<0.05)。中、高剂量组雄鼠体质量从染毒第14天开始均低于对照组和低剂量组雄鼠(P<0.05)。高剂量组孕鼠体质量在妊娠第0、7、14、19天均低于对照组、低剂量组和中剂量组(P<0.05);中剂量组孕鼠体质量在妊娠第0天低于低剂量组(P<0.05),在妊娠第7、14天均低于对照组和低剂量组(P<0.05)。高剂量组雌鼠的受孕率和仔鼠出生存活率、哺乳成活率均低于其余3组雌鼠(P<0.008);中剂量组雌鼠的仔鼠哺乳成活率分别低于对照组和低剂量组(P<0.008)。中、高剂量组雌鼠脑脏器系数均高于对照组和低剂量组雌鼠(P<0.05);中、高剂量组雄鼠脑、睾丸脏器系数均高于对照组和低剂量组雄鼠(P<0.05),高剂量组雄鼠附睾脏器系数均低于其余3组雄鼠(P<0.05)。(2)子一代。子一代低、中剂量组雌鼠在染毒第42天体质量高于对照组雌鼠(P<0.05);低剂量组雄鼠在染毒第42、49、56天体质量均高于对照组雄鼠(P<0.05);中剂量组雄鼠在染毒第14、21、42、49、56天体质量均高于对照组(P<0.05)。低剂量组仔鼠出生存活率低于对照组(P<0.017);高剂量组仔鼠出� Objective To obtain information on the toxicity of lufenuron on the reproduction ability and the growth and development of offspring in female and male rats through two-generation reproduction toxicity study.Methods The specific pathogen free healthy SD rats were randomly divided into control group and low-,medium-and high-dose lufenuron groups,with 60 rats in each group,half females and half males.Rats in the low-,medium-and high-dose lufenuron groups were respectively fed with lufenuron at the dose of 5.0,20.0 and 80.0 mg/(kg body weight·day)for 8 weeks before mating.The control group was fed with standard foot.The reproductive index,brain and reproductive organ coefficients and pathological changes were observed in P and F1 parents.The birth and growth indexes of the offspring were measured.Results i)P generation:from the 14 th day,the female rats in the medium-dose group had lower body weight than that of the female control group(P<0.05);from the 35 th day,the body weight was lower than that of the female low-dose group(P<0.05).From the 14 th day,the female rats in the high-dose group had lower body weight than that of the other three female groups(P<0.05).From the 14 th day,the male rats in the medium-and high-dose groups had lower body weight than that of the male control group and low-dose group(P<0.05).The body weight of pregnant rats in the parental high-dose group was lower than that of the control group,low-dose group,and medium-dose group at day 0,7,14,19 of the pregnancy duration(P<0.05).The body weight of pregnant rats in the parental medium-dose group was lower than that of the low-dose group on day 0 of the pregnancy duration,and lower than that of the control and low-dose groups on day 7 and 14(P<0.05).The conception rate,the new-borne survival rates and the feeding survival rate of female rats in the high-dose group was lower than that of the other three female groups(P<0.008).The new-borne feeding survival rate of female rats in the medium-dose group was lower than that of the control group an
