摘要
阐明藏猪和长白猪心脏血管铸型结构差异,为揭示藏猪适应高海拔低氧环境的生物学机制提供理论参考。采集甘南藏猪、移居至泾川的藏猪、移居到永登的长白猪和泾川长白猪等不同生长环境下的心脏组织,采用ABS[丙烯腈(A)、丁二烯(B)、苯乙烯(S)三种单体的三元共聚物]灌注剂制作血管铸型,并通过扫描电镜观察分析心脏血管铸型特征。结果显示,藏猪心脏前室间支的内侧血管较长白猪发达,且枝干上有较多小动脉,长白猪前室间支内径与藏猪相比差异显著(P<0.05);藏猪心脏大静脉血管走形较为弯曲且伸展覆盖整个铸型面积较长白猪广,长白猪心大静脉内径与藏猪相比差异显著(P<0.05);通过分析不同群体心脏前室间支及心大静脉管径与心脏质量的相对值(T/W)发现,藏猪群体T/W值均显著高于长白猪群体(P<0.05),较高海拔的甘南藏猪群体、永登长白猪群体的T/W值均分别高于较低海拔的泾川藏猪群体和泾川长白猪群体,但差异均不显著。扫描电镜观察发现,长白猪心脏微血管铸型表面分布有较浅的平滑肌纤维压痕,偶尔可见卵圆形的内皮细胞核压痕,而藏猪心脏微血管铸型表面呈现"环形缩窄"与内皮细胞核压痕伴行特征。说明同长白猪相比,藏猪对血流的调节能力更强,拥有更强的供血能力,这可能是藏猪为适应高海拔环境长期进化的结果。
To explore the differences in the structure of heart vessels in Tibetan pigs and Landrace pigs,and to provide a theoretical basis for the impact of high altitude environment on Tibetan pigs.anterior interventricular branch of the Tibetan pig are longer and the white pigs are developed.In this study,we collected the heart tissue of Gannan Tibetan pig,Jingchuan Tibetan pig,Yongdeng Landrace pig and Jingchuan Landrace pig,and made blood vessel casts with ABS perfusion agent,and observed and analyzed the characteristics of blood vessel casts by SEM.The results showed that the medial vessels of the anterior interventricular branch of Tibetan pig were more developed than that of Landrace pig,and there were more small arteries on the branches,and the diameter of the anterior interventricular branch of Landrace pig was significantly different from that of Tibetan pig(P<0.05);the shape of the great vein of Tibetan Pig was more curved and extended to cover the whole casting area,and the diameter of the great vein of Landrace pig was significantly different from that of Tibetan pig(P<0.05)The results showed that the T/W value of Tibetan pig population was significantly higher than that of Landrace group(P<0.05),and the T/W value of Gannan Tibetan pig population and Yongdeng Landrace group were higher than that of Jingchuan Tibetan pig population and Landrace group,but the difference was not significant.The results of scanning electron microscopy showed that there were shallow smooth muscle fiber indentation on the surface of the heart microvascular cast of Landrace pig,occasionally oval endothelial cell nuclear indentation,while the surface of the heart microvascular cast of Tibetan pig showed the characteristics of"ring narrowing"and endothelial cell nuclear indentation.It was found that compared with Landrace pigs,Tibetan pigs have a stronger ability of blood flow regulation and blood supply,which may be the result of long-term evolution of Tibetan pigs to adapt to high altitude environment.
