摘要
目的探讨脐血干细胞移植后帕金森病(Parkinson’s disease,PD)大鼠旋转行为和肿瘤坏死因子α(tumor necrosis factorα,TNF-α)及白细胞介素-4(interleukin-4,IL-4)的变化。方法将20只PD大鼠模型随机分为对照组和实验组,各10只,对照组在纹状体植入点注射5μL磷酸盐缓冲溶液(PBS),实验组注射5μL脐血来源间充质干细胞(human umbilical cord blood-derived mesenchymal stem cell,HUMSCs)(浓度为3.0×105μL-1)。移植前和移植后3、6、9周分别观察大鼠旋转行为的改变,用酶联免疫吸附测定法(ELISA)检测TNF-α及IL-4的浓度。结果与对照组比较,实验组在第3周旋转行为、TNF-α及IL-4水平的改变差异无统计学意义(P>0.05);在移植后6、9周,旋转行为分别减少为(7.80±1.90)r·min-1、(5.60±0.40)r·min-1,TNF-α分别降低为(88.36±11.29)ng·L-1、(69.69±10.50)ng·L-1,IL-4分别增高为(906.00±16.29)ng·L-1、(984.39±42.50)ng·L-1,差异均有统计学意义(P<0.05)。与移植前比较,实验组在第3周各指标的改变差异无统计学意义(P>0.05);在移植后6、9周,旋转行为、TNF-α及IL-4水平的改变差异有统计学意义(P<0.05)。结论HUMSCs移植后一定时间可以明显改善PD大鼠旋转行为,可以显著减低TNF-α水平和提高抗炎因子IL-4的水平。
Objective To investigate the changes of rotation behavior,tumor necrosis factorαand interleukin-4 in Parkinson’s disease(PD)rats after umbilical cord blood stem cell transplantation.Methods 20 PD rats were randomly divided into the control group(n=10)and experimental group(n=10).The control group was injected with 5μL phosphate buffered solution(PBS).The experimental group was injected with 5μL human umbilical cord blood-derived mesenchymal stem cell(HUMSCs)(concentrstion:3.0×105μL-1).The changes of rotational behavior were observed before transplantation and at the 3,6,9 weeks after transplantation.The concentrations of tumor necrosis factor a tumor necrosis factorα(TNF-α)and interleukin-4(IL-4)were measured by enzyme linked immunosorbent assay(ELISA).Results Compared with the control group,there was no significant difference in rotational behavior,TNF-αand IL-4 levels in the experimental group at the 3rd week(P>0.05).At the 6 and 9 week,the rotation behavior decreased to(7.80±1.90)r·min-1 and(5.60±0.40)r·min-1 respectively;TNF-αdecreased to(88.36±11.29)ng·L-1 and(69.69±10.50)ng·L-1 respectively;IL-4 increased to(906.00±16.29)ng·L-1 and(984.39±42.50)ng·L-1 respectively,the differences were statistically significant(P<0.05).Compared with pre-transplantation,there was no significant change in every index in the experimental group at the 3rd week(P>0.05),but there was a significant difference in the 6th and 9th week in rotational behavior,TNF-αand IL-4 levels(P<0.05).Conclusion At a certain time after transplantation of HUMSCs,the rotation behavior of PD rats was significantly improved,TNF-αlevel significantly decreased and anti-inflammatory factor IL-4 level was significantly increased.
