摘要
目的研究超高效液相色谱(UPLC)法在测定黄芪降压方提取液中黄芪甲苷、柚皮苷两种活性成分的含量。方法利用UPLC对黄芪降压方的提取药液进行分析,流动相用乙腈-去离子水测定,色谱柱运用Agilent SB C18柱来测定,ESI离子源的检测离子对分别是829/783及579/271,以此实施UPLC测定,分析UPLC的应用价值。结果黄芪甲苷、柚皮苷线性关系较好(r>0.999);黄芪甲苷平均回收率为100.34%,RSD值为1.89%;柚皮苷平均回收率为101.02%,RSD值为1.74%;连续6次测定,黄芪甲苷RSD值为1.36%,柚皮苷为1.43%;不同人员进行检测,黄芪甲苷RSD值为1.80%,柚皮苷为2.27%,不同时间测定,黄芪甲苷RSD值为2.55%,柚皮苷为1.91%;24 h内黄芪甲苷的RSD值为1.85%,柚皮苷的为2.97%。结论 UPLC对于黄芪降压方中黄芪甲苷、柚皮苷检测具有较高准确性和精密度,可为临床评价治疗效果质量提供参考数据。
Objective To study the content of astragaloside Ⅳ and naringin in the extract of Astragalus hypotension prescription tested by ultra high performance liquid chromatography(UPLC). Methods The extracted liquid of Astragalus hypotensive prescription was analyzed by UPLC, the mobile phase was tested by acetonitrile-deionized water, the chromatographic column was tested by Agilent SB C18. The detection ion pair of ESI ion source was 829/783 and 579/271, respectively, they were tested by UPLC to analyze the application value of UPLC. Results The linear relationship between astragaloside Ⅳ and naringin was good(r > 0.999). The average recovery rate of astragaloside Ⅳ was 100.34% and RSD value was 1.89%. The average recovery rate of naringin was 101.02% and RSD value was 1.74%. 6 consecutive tests showed that the RSD value of astragaloside Ⅳ and naringin were 1.36% and 1.43%, respectively. Tests by different persons showed that the RSD value of astragaloside Ⅳ and naringin were 1.80% and 2.27%, respectively. Tests in different times point showed that the RSD value of astragaloside Ⅳ and naringin were 2.55% and 1.91%, respectively. Within 24 h, the RSD value of astragaloside Ⅳ and naringin were 1.85% and 2.97%, respectively. Conclusion UPLC has high accuracy and precision in detecting astragaloside Ⅳ and naringin in Astragalus hypotensive prescription, which can provide reference data for clinical evaluation of therapeutic effect quality.
