摘要
目的针对益气生血胶囊中黄芪甲苷进行含量检测,建立相应的反相高效液相色谱(RP-HPLC)方法。方法色谱柱为Agilent Eclipse XDB-C18柱(150 mm×3.9 mm,5μm),流动相为乙腈-0.1磷酸溶液(30∶70),流速为1 mL/min,柱温为30℃,紫外检测波长为207 nm;进样量为25μL。结果在0.01 mg/mL、0.05 mg/mL、0.2 mg/mL、0.5 mg/mL、1 mg/mL及2 mg/mL线性范围内,黄芪甲苷均与峰面积呈良好的线性关系,其精密度、稳定性及重复性均可,均回收率值为96.5%,平均RSD值为2.38%。结论所建立的反相高效液相色谱法,应用于益气生血胶囊中黄芪甲苷的测定,准确性高,重复性好,且操作简便,可作为益气生血胶囊的质量控制标准。
Objective To establish a corresponding RP-HPLC method for the content determination of Astragaloside IV in Yiqi Shengxue capsule.Methods The chromatographic column was Agilent Eclipse XDB-C18(150 mm×3.9 mm,5μm),the mobile phase was acetonitrile-0.1 phosphoric acid solution(30∶70),the flow rate was 1 mL/min,the column temperature was 30℃,and the UV detection wavelength was 207 nm.The injection volume was 25μL.Results Within the linear range of 0.01 mg/mL,0.05 mg/mL,0.2 mg/mL,0.5 mg/mL,1 mg/mL and 2 mg/mL,there was a good linear relationship between the peak area and astragaloside IV.The precision,stability and repeatability were all good.The average recovery value was 96.5%and the average RSD value was 2.38%.Conclusion The RP-HPLC method is applied to the determination of Astragaloside IV in Yiqi Shengxue capsule with high accuracy,good repeatability and simple operation,which can be used as the quality control standard of Yiqi Shengxue capsule.
