摘要
目的研究卵巢癌细胞中NNT-AS1的表达情况,探索NNT-AS1沉默后对卵巢癌细胞增殖、凋亡及转移的影响。方法使用qRT-PCR检测卵巢癌HO-8910、SK-OV-3细胞的NNT-AS1表达情况。随后将卵巢癌HO-8910、SK-OV-3细胞分别分为siNNT-AS1组(NNT-AS1沉默组)、siNC组(阴性对照组)和control组(对照组),siNNT-AS1组和siNC组分别转染NNT-AS1沉默质粒siNNT-AS1和阴性对照质粒siNC,control组给予空载体。72 h后采用MTT法检测细胞增殖活性,流式细胞术检测细胞凋亡,Transwell实验检测细胞侵袭能力,并采用比色法检测caspase-3、caspase-9活力。结果卵巢癌HO-8910、SK-OV-3细胞中NNT-AS1表达均低于正常卵巢细胞T29,差异有统计学意义(P<0.05)。与siNC组及control组HO-8910细胞系、SK-OV-3细胞系相比,siNNT-AS1组24,48,72 h的细胞增殖活性均增高(均P<0.05),细胞凋亡率均下降(均P<0.05),细胞侵袭能力均升高(均P<0.05),caspase-3、caspase-9活性均降低(均P<0.05)。结论NNT-AS1在人卵巢癌HO-8910、SK-OV-3细胞中表达降低,NNT-AS1表达下降可能与卵巢癌细胞的增殖和侵袭增加及凋亡下降有关;NNT-AS1可能通过抑制内源性途径进而调控卵巢癌细胞凋亡。
Objective To study the expression of NNT-AS1 in ovarian cancer cells,and to explore the effect of NNT-AS1 silencing on the proliferation,the apoptosis and the invasion of ovarian cancer cells.Methods The expression of NNT-AS1 in ovarian cancer cells such as HO-8910 and SK-OV-3 was detected by qRT-PCR.Then HO-8910 and SK-OV-3 cells were respectively divided into siNNT-AS1 group(NNT-AS1 silencing group),siNC group(negative control group)and control group.HO-8910 and SK-OV-3 cells were respectively transfected with NNT-AS1 silencing plasmid siNNT-AS1 and negative control plasmid siNC in siNNT-AS1 group and siNC group,and empty vector was given in control group.After 72 h,the cell proliferation activity was detected by MTT assay,the cell apoptosis was detected by flow cytometry,the cell invasion ability was detected by Transwell assay,and caspase-3 and caspase-9 activities were detected by colorimetry.Results The expression of NNT-AS1 in ovarian cancer cells HO-8910 and SK-OV-3 was statistically lower than that in normal ovarian cell line T29(P<0.05).Compared with siNC group and control group,the cell proliferation activities of HO-8910 cells and SK-OV-3 cells were increased in siNNT-AS1 group at 24 h,48 h and 72 h(all P<0.05),the cell apoptosis rats were decreased(all P<0.05),the cell invasion abilities were increased(all P<0.05),and the caspase-3 and caspase-9 activities were decreased(all P<0.05).Conclusion NNT-AS1 expression decreases in human ovarian cancer cells such as HO-8910 and SK-OV-3.The decrease of NNT-AS1 expression in ovarian cancer cells may be related to the increase of proliferation and invasion and the decrease of apoptosis.NNT-AS1 may regulate the apoptosis of ovarian cancer cells by inhibiting the endogenous pathway.
