摘要
为研究右美托咪定(dexmedetomidine,Dex)联合顺铂对Lewis肺癌小鼠免疫功能的影响,将72只C57BL/6J小鼠随机分为6组(每组12只)如下:对照组(N组)、假手术组(S组)、模型组(M组)、Dex组(D组)、顺铂组(C组)及Dex联合顺铂组(DC组)。称量小鼠瘤体积、瘤质量、体质量及脾脏、胸腺质量;MTT法检测肿瘤细胞增殖活性;FCM检测肿瘤细胞凋亡率;ELISA检测小鼠血清TNF-α、IL-6表达水平;Western blotting检测肿瘤组织中IL-6、信号转导与转录激活因子3(signal transducer and activator of transcription 3, STAT3)、p-STAT3蛋白表达量。结果显示,DC组小鼠瘤质量、瘤体积、瘤组织IL-6水平、p-STAT3/STAT3蛋白表达量均显著低于D、C组(P<0.05),肿瘤细胞凋亡率、脾脏、胸腺指数、白细胞(white blood cell,WBC)、骨髓细胞(bone marrow cell,BMC)、淋巴细胞计数、淋巴细胞增殖活性、血清TNF-α水平均显著高于D、C组(P<0.05)。由此,Dex联合顺铂可促进Lewis肺癌小鼠免疫功能,减轻化疗对小鼠的免疫抑制,可能与抑制IL-6/STAT3信号通路激活有关。
To explore the effect of dexmedetomidine(Dex) combined with cisplatin on immune function of Lewis lung cancer mouse, 72 C57 BL/6 J mice were randomly divided into 6 groups(12 mice per group): control group(group N), sham operation group(group S), model group(group M), Dex group(group D), cisplatin group(group C) and Dex combined with cisplatin group(group DC). The tumor volume, weight, body weight and thymus, spleen weight were measured;The tumor cell proliferation rate was detected by MTT;The apoptosis rate of tumor cells was detected by FCM;the expressions of TNF-α and IL-6 in serum were detected by ELISA;the protein expressions of IL-6, signal transducer and activator of transcription 3(STAT3) and p-STAT3 in tumor tissues were detected by Western blotting. The results showed that tumor weight, tumor volume, serum IL-6 levels, and p-STAT3/STAT3 protein expressions in tumor tissue of DC group were lower than those in D and C group(P<0.05), the apoptosis rate of tumor cells, spleen, thymus index, white blood cell(WBC), bone marrow cell(BMC), lymphocyte count, lymphocyte proliferation activity and serum TNF-α levels in DC group were higher than those in group D and C(P<0.05). Therefore, Dex combined with cisplatin can promote the immune function of Lewis lung cancer mice and reduce the immunosuppression induced by cisplatin, which may be related to the inhibition of IL-6/STAT3 pathway activation.
