白藜芦醇改善妊娠期糖尿病大鼠及子代糖耐量和胰岛功能 被引量:5

Resveratrol Improves Glucose Tolerance and Islet Function in Gestational Diabetic Rats and Their Offspring

摘要 目的探讨白藜芦醇(RESV)对妊娠期糖尿病(gestational diabetes mellitus,GDM)大鼠及其子鼠糖耐量和胰岛功能的作用。方法采用饮食诱导肥胖的妊娠期糖尿病大鼠,在孕晚期给予RESV膳食补充,检测其糖耐量水平和胰岛功能。其子鼠分别给予低脂饮食和高脂饮食,在15周龄时检测其糖耐量水平和胰岛功能,免疫荧光检测其胰岛中胰岛素水平。结果与对照组比较,GDM组、GDM+RESV组母鼠的进食量降低,体质量增加(P均<0.05),且GDM+RESV组母鼠体质量低于GDM组(P<0.05)。与对照组高脂饮食子鼠相比,GDM和GDM+RESV组高脂饮食子鼠体质量增加,且GDM+RESV组较GDM组体质量低(P均<0.05)。GDM组母鼠血糖浓度与糖耐量高于对照组(P<0.05),且GDM+RESV组低于GDM组(P<0.05)。与对照组母鼠相比,GDM组、GDM+RESV组母鼠血浆胰岛素浓度、HOMA-IR指数和离体总胰岛素含量均下降(P均<0.05),且GDM+RESV组高于GDM组(P<0.05)。GDM组子鼠的血糖浓度与糖耐量水平高于对照组子鼠(P<0.05),且GDM+RESV组子鼠的血糖浓度与糖耐量水平低于GDM组子鼠(P<0.05)。与对照组相比,GDM组子鼠的血浆胰岛素浓度、离体胰高血糖素分泌量及离体总胰高血糖素含量均降低,离体胰岛素分泌量升高(P均<0.05),且GDM+RESV组血浆胰岛素浓度、离体胰高血糖素分泌量、离体总胰岛素含量均升高,离体胰岛素分泌量低于GDM组(P均<0.05)。免疫荧光检测显示,与对照组相比,GDM组子鼠的胰岛素水平减弱,而GDM+RESV组子鼠的胰岛素水平升高。结论GDM大鼠及其子鼠糖耐量和胰岛功能均降低,膳食补充RESV可改善GMD及其子鼠糖耐量和胰岛功能。 Objective To explore the effects of resveratrol(RESV)on glucose tolerance and islet function in gestational diabetic rats and their offspring.Methods Diet-induced obese gestational diabetic rats were given RESV dietary supplement in the third trimester to detect their glucose tolerance and islet function.The offspring were given a low-fat diet and a high-fat diet,and their glucose tolerance and pancreatic islet function were detected at 15 weeks of age.The insulin levels in their pancreatic islets were detected by immunofluorescence.Results Compared with the control group,the food intake of the mother mice in the GDM group and the GDM+RESV group decreased,and the body mass increased(P all<0.05),and the body mass of the mother mice in the GDM+RESV group was lower than that of the GDM group(P<0.05).Compared with the control group high-fat diet mice,the body mass of the high-fat diet mice of the GDM and GDM+RESV groups increased,and the body mass of the GDM+RESV group were lower those that of the GDM group(P all<0.05).The blood glucose concentration and glucose tolerance of female rats in the GDM group were higher than those in the control group(P<0.05),and the GDM+RESV group was lower than the GDM group(P<0.05).Compared with the control group,the plasma insulin concentration,HOMA-IR index and total insulin content of the GDM group and the GDM+RESV group were all decreased(P all<0.05),and the GDM+RESV group was higher than the GDM group(P<0.05).The blood glucose concentration and glucose tolerance level of the offspring of the GDM group were higher than those of the control group(P<0.05),and the blood glucose concentration and glucose tolerance level of the offspring of the GDM+RESV group were lower than those of the GDM group(P<0.05).Compared with the control group,the plasma insulin concentration,isolated glucagon secretion and total glucagon content of the offspring of the GDM group decreased,and the isolated insulin secretion increased(P all<0.05),and plasma insulin concentration,isolated glucagon secretion,and
