摘要
[目的]观察己酮可可碱(PTX)对神经病理性疼痛小鼠海马中p-p38,p-JNK蛋白的表达及抑郁行为的影响.[方法]利用坐骨神经套扎术(Cuff)建立神经病理性疼痛小鼠模型,通过小鼠后肢回缩压力阈值测定(PWPT)、蔗糖偏好试验、旷场试验等行为学检测方法评估Cuff诱导的神经病理性疼痛小鼠神经病理性疼痛症状和抑郁样行为;采用ELISA法检测小鼠血清中TNF-α,IL-6水平,Western blot法检测海马组织中p-p38和p-JNK蛋白的表达情况.[结果]与Cuff组比较,Cuff+PTX组PWPT值、蔗糖消耗比值、水平得分及垂直得分均显著升高(P<0.05),TNF-α,IL-6水平均明显降低(P<0.05),p-JNK,p-p38表达均明显下调(P<0.05).[结论] PTX可能通过抑制p38和JNK活性及降低炎症因子而缓解Cuff所致的小鼠抑郁样行为.
OBJECTIVE To observe the effects of pentoxifylline(PTX) on the expressions of p-p38 and p-JNK and depression-like behaviors in the hippocampus of neuropathic pain mice. METHODS A mouse model of neuropathic pain was established by sciatic nerve cuffing(Cuff). Neuropathic pain symptoms and depression-like behaviors induced by Cuff in mice with neuropathic pain were evaluated with paw withdrawal pressure threshold test(PWPT), sucrose preference test(SPT) and open field test(OFT) and other behavioral tests. ELISA was used to detect the levels of TNF-α and IL-6 in serum of the mice. Western blot was used to detect the expressions of the p-p38 and p-JNK in the hippocampus. RESULTS Compared with the Cuff group, PWPT value, sucrose consumption ratio, horizontal and vertical score in the Cuff + PTX group were significantly increased(P<0.05), the levels of TNF-α and IL-6 were significantly decreased(P<0.05), the expressions of p-p38 and p-JNK were significantly downregulated(P<0.05). CONCLUSION PTX might alleviate depression-like behavior in mice induced by Cuff by inhibiting p38 and JNK activity and reducing inflammatory factors.
