摘要
目的探讨ILC2、Th2细胞在香烟烟雾暴露的变应性鼻炎大鼠模型中的变化。方法将15只大鼠分为正常组(Normal)、变应性鼻炎组(AR)、烟雾暴露组(Somke+AR),每组5只。造模后对各组大鼠进行AR症状评分,苏木精-伊红(HE)和过碘酸雪夫(PAS)染色法观察大鼠鼻黏膜炎症情况;酶联免疫吸附试验(ELISA)检测外周血总IgE、IL-10、IL-13水平;流式细胞术检测脾脏中ILC2、Th2细胞比例变化;免疫组化法检测大鼠鼻黏膜GATA3、RORα表达水平。多组间比较采用单因素方差分析,两两比较采用LSD-t检验。结果AR组大鼠的过敏症状、鼻黏膜嗜酸粒细胞浸润及血清总IgE水平较Normal组升高,而Smoke+AR组较AR组进一步升高。探讨机制发现,与Normal组(0.82±0.69、0.98±0.24)比较,AR组(3.70±0.78、2.05±0.26)和Smoke+AR组(7.72±0.90、3.46±1.08)ILC2、Th2细胞表达均升高(P<0.05),其中Smoke+AR组升高最明显。ELISA法检测Th2细胞因子显示,与Normal组(15.96±4.39、45.33±10.07)比较,IL-10、IL-13表达水平在AR组(33.88±5.73、336.00±100.00)升高(P<0.05),在Smoke+AR组(86.38±14.42、636.00±190.79)进一步升高(P<0.05)。免疫组化法检测大鼠鼻黏膜显示与Normal组(1.04±0.18、0.48±0.28)比较,GATA3、RORa表达水平在AR组(3.55±0.79、1.88±0.45)和Smoke+AR组(6.13±0.87、3.61±0.75)逐渐升高(P<0.05),其中Smoke+AR组升高明显。结论香烟烟雾暴露可以促进变应性鼻炎大鼠的变应性炎症反应,其机制可能与促进ILC2和Th2细胞分化及其细胞因子分泌增加有关。
Objective To investigate the changes of ILC2 and Th2 cells in rat model of allergic rhinitis which exposed to cigarette smoke.Methods Fifteen rats were divided into 3 groups:Normal group(n=5),AR group(n=5),and Smoke+AR(n=5).Symptoms of allergic rhinitis were scored after the establishment of rat model in each group.HE and PAS staining methods were used to observe the inflammatory infiltration in nasal mucosa.ELISA method was used to detect the serum levels of total IgE,IL-10,and IL-13.Flow cytometry was used to detect the change of ILC2 and Th2 cells.Immunohistochemical staining was used to detect the expression of GATA3 and retinoic acid receptor associated solitary nucleus receptor(ROR)in rat nasal mucosa.Multiple group and two group comparisons were performed by using a one-way analysis of variance and LSDt test respectively.Results The allergic symptoms,eosinophilic infiltration of nasal mucosa and total serum IgE level in AR group were higher than those in Normal group,and Smoke+AR group were higher than AR group.It was further found that compared with Normal group(0.82±0.69,0.98±0.24),the expression of ILC2 and Th2 cells in AR group(3.70±0.78,2.05±0.26)and Smoke+AR group(7.72±0.90,3.46±1.08)increased(P<0.05),especially in Smoke+AR group.ELISA method was used to detect the expression of Th2 cytokines,and compared with Normal group(15.96±4.39,45.33±10.07),the expression level of IL-10 and IL-13 increased in AR group(33.88±5.73,336.00±100.00)(P<0.05),and further increased in Smoke+AR group(86.38±14.42,636.00±190.79)(P<0.05).Immunohistochemical method was used to detect the expression of rat nasal mucosa,and compared with Normal group(1.04±0.18,0.48±0.28),the expression level of GATA3 and RORa increased gradually in AR group(3.55±0.79,1.88±0.45)and Smoke+AR group(6.13±0.87,3.61±0.75)(P<0.05),especially in Smoke+AR group.Conclusion Cigarette smoke exposure can promote allergic inflammatory responses in rats with allergic rhinitis,which may be related to promotion of ILC2 and Th2 cell differenti
