摘要
目的:研究不同质量浓度的氮酮对蟾酥凝胶贴膏体外透皮效果的影响及蟾酥有效成分的透皮吸收特性。方法:采用改良Franz装置,以雄性昆明种小鼠腹部皮肤作为供试皮肤,用高效液相色谱法(HPLC)测定不同质量浓度(0%,1%,3%,5%)的氮酮对蟾酥有效成分渗透量、吸收速度的影响。结果:氮酮能够显著增加蟾酥有效成分的累积渗透量和渗透速率,且效果与氮酮含量呈正比;经透皮后华蟾酥毒基与脂蟾毒配基在皮肤中均有一定的蓄积。结论:氮酮质量浓度为5%时华蟾酥毒基与脂蟾毒配基体外透皮效果最好。
Objective:To study the effect of different mass concentrations of azone on transdermal absorption of toad venom gel plaster in vitro and the transdermal absorption characteristics of effective ingredients in toad venom. Methods:The modified Franz device and high-performance liquid chromatography(HPLC) were used to determine the effect of different mass concentrations(0%,1%,3%,5%) of azone on the penetration rate and absorption rate of effective ingredients of toad venom with the abdominal skin of male Kunming mice as the test skin. Results:Azone could significantly increase the cumulative penetration rate and penetration rate of effective ingredients of toad venom,and the effect was proportional to the content of azone.After transdermal absorption,there were certain accumulations in the skin with cinobufagin and resibufogenin. Conclusions:When the mass concentration of azone was 5%,the transdermal absorption effect of cinobufagin and resibufogenin in vitro achieved the best.
