摘要
目的:探讨白藜芦醇对SD大鼠脑组织缺血再灌注过程中炎症通路及炎性坏死的影响,深入认识白藜芦醇改善脑梗死的分子机制。方法:采用随机数表法将选取的40只SD大鼠分为假手术组(8只)、模型组(8只)和白藜芦醇组[白藜芦醇组又分为低剂量组(20 mg/kg,8只)、中剂量组(50 mg/kg,8只)和高剂量组(100 mg/kg,8只)]。造模成功后检测白藜芦醇干预后SD大鼠脑组织的梗死面积,选择最佳白藜芦醇干预剂量;检测SD大鼠血浆中乳酸脱氢酶(LDH)活性、丙二醛(MDA)及相关炎性因子水平;检测肿瘤坏死因子-α(TNFα)和白介素-1β(IL-1β)水平,检测toll样受体4(TLR4)和核转录因子-κB(NF-κB)(TLR4/NF-κB)通路蛋白的激活水平表达。结果:SD大鼠脑组织缺血再灌注后,模型组的梗死面积显著高于白藜芦醇低剂量组和中剂量组,其差异有统计学意义(t=1.88,t=2.52;P<0.05);而白藜芦醇高剂量组与白藜芦醇中剂量组SD大鼠的脑梗死面积比较无显著改善。三组mRNA的相关基因TNFα、IL-1β和Caspase-1水平比较,差异有统计学意义(F=408.723,F=900.339,F=541.870;P<0.05),三组LDH活性和MDA含量有统计学意义(F=494.310,F=152.358;P<0.05)。结论:白藜芦醇可能抑制TLR4/NF-κB通路蛋白下调炎性细胞坏死相关因子的表达,对脑组织缺血再灌注损伤具有一定保护作用。
Objective:To investigate the effect of resveratrol on inflammatory pathways and inflammatory necrosis in the process of ischemia reperfusion of brain tissue of SD rat so as to deeply recognize the molecular mechanism of resveratrol in improving cerebral infarction.Methods:SD rats were randomly divided into sham operation group,model group,low dose group of resveratrol(20 mg/kg),middle dose group of resveratrol(50 mg/kg)and high dose group of resveratrol(100 mg/kg)by random number method.After the model was established,the infarct size of brain tissue of SD rat with the intervention of resveratrol was detected,and then the optimal intervention dose of resveratrol was selected.The activity of plasma lactic dehydrogenase(LDH),malondialdehyde(MDA)and the levels of relevant inflammatory factors in SD rats were detected.Besides,the levels of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β),and the expression of activation level of pathway protein of toll-like receptor 4(TLR4)and nuclear transcription factor-kB(NF-κB)(TLR4/NF-κB)were detected.Results:After the brain tissue of SD rat was ischemia reperfusion,the infarct size of model group was significantly higher than that of low dose group of resveratrol(t=1.88,P<0.05)and middle dose group of resveratrol(t=2.52,P<0.05).While compared with that in middle dose group,the cerebral infarction size of SD rats in high dose group of resveratrol wasn’t significantly improved.The differences of the relevant gene TNF-α,IL-1βand Caspase-1 of mRNA among three groups were significant(F=408.723,F=900.339,F=541.870,P<0.05).And the differences of LDH-activity and the MDA content among three groups were significant(F=494.310,F=152.358,P<0.05).Conclusion:Resveratrol can restrain the expression of the relevant factors of inflammatory necrocytosis of protein down-regulation in the TLR4/NF-κB pathway,and it has a certain protective effect on ischemia-reperfusion injury of brain tissue in SD rats.
