二氢杨梅素对结肠炎小鼠炎性因子及结肠组织核因子-κB p65表达的影响

Effect of dihydrobayberry on the inflammatory cytokines and colon tissues nuclear factor-κB p65 expression of colitis mice

摘要 目的研究二氢杨梅素对结肠炎小鼠炎性因子及结肠组织核因子-κB p65(NF-κB p65)表达的影响。方法将45只SPF级BALB/c小鼠随机分为对照组、模型组及实验组,各15只。模型组及实验组小鼠连续饮用5%葡聚糖硫酸钠溶液7 d构建溃疡性结肠炎小鼠模型,对照组饮用蒸馏水。实验组建模期间灌胃200 mg·kg^-1·d^-1二氢杨梅素,对照组与模型组则灌胃等量生理盐水,每天1次,连续干预2周。以酶联免疫吸附(ELISA)法检测血清炎性因子含量;以苏木精-伊红(HE)染色观察小鼠结肠组织病理学形态;以蛋白质印迹(WB)法检测结肠组织NF-κB p65表达。结果对照组、模型组及实验组小鼠血清白细胞介素-1β(IL^-1β)含量分别为(42.11±5.32),(257.03±11.76)和(152.17±12.14)pg·mL^-1;白细胞介素-2(IL-2)含量分别为(30.05±4.79),(54.18±5.02)和(41.23±4.81)ng·L^-1;白细胞介素-6(IL-6)含量分别为(102.38±17.62),(469.15±48.25),(399.58±45.16)pg·mL^-1;肿瘤坏死因子-α(TNF-α)含量分别为(76.42±11.21),(296.03±33.42)和(201.56±23.78)pg·mL^-1;结肠组织学评分分别为(0.81±0.22),(6.02±0.45)和(3.64±0.33)分。模型组血清IL^-1β、IL-2、IL-6、TNF-α含量,结肠组织学评分及NF-κB p65、p-NF-κB p65蛋白表达量比对照组高,实验组比模型组低,差异均有统计学意义(均P<0.05)。结论二氢杨梅素可有效抑制结肠炎小鼠炎性反应,改善结肠组织炎性病理损伤,其作用机制可能与有效下调结肠组织NF-κB p65与p-NF-κB p65表达相关。 Objective To explore the effect of dihydrobayberry on the inflammatory cytokines and colon tissues nuclear factor-κB p65(NF-κB p65)expression of colitis mice.Methods Forty-five SPF grade BALB/c mice were randomly divided into control group,model group and test group,15 mice in each group.Model group and test group were built ulcerative colitis mice model by drinking 5%dextran sodium sulfate solution for 7 d,control group was given distilled water.During the modeling period,test group was given 200 mg·kg^-1·d^-1 dihydromyricetin by gavage,control group and model group were given the same amount of normal saline by gavage,once a day for 2 weeks.The serum inflammatory factor contents were detected by enzyme-linked immunosorbent experiment(ELISA);the mice colon tissues pathology morphology was observed by hematoxylin and eosin(HE)staining;the expression of NF-κB p65 was detected by Western blot(WB)method.Results The serum interleukin-1β(IL^-1β)in control group,model group and test group were(42.11±5.32),(257.03±11.76),(152.17±12.14)pg·mL^-1;the serum interleukin-2(IL-2)were(30.05±4.79),(54.18±5.02),(41.23±4.81)ng·L^-1;the serum interleukin-6(IL-6)were(102.38±17.62),(469.15±48.25),(399.58±45.16)pg·mL^-1;the serum tumor necrosis factor-α(TNF-α)contents were(76.42±11.21),(296.03±33.42),(201.56±23.78)pg·mL^-1;colonic histological grading were(0.81±0.22),(6.02±0.45),(3.64±0.33)scores.The serum IL^-1β,IL-2,IL-6,TNF-αcontents,the colon histological score and NF-κB p65,p-NF-κB p65 protein expression in model group were higher than those in control group,test group were lower than model group,all with significant differences(all P<0.05).Conclusion Dihydrobayberry can inhibit the colitis mice inflammatory reaction effectively and improve the colon tissues inflammatory pathological damage,the action mechanism may be related to the significant down-regulation of the expressions of colon tissues NF-κB p65 and phosphorylation-NF-κB p65.
