草决明提取物对大鼠脑外伤的保护作用 被引量:3

摘要 目的:观察草决明提取物对脑外伤大鼠中枢神经损伤的保护作用。方法:随机将72只大鼠分为假手术组、脑外伤组和草决明(Cassia o btusifolial)提取物高(COB 20 g/kg)、低(COB10 g/kg)剂量组,除假手术组外,其他组均以Feeney’s自由落体法制备脑外伤模型,而假手术组仅做颅骨开窗,草决明提取物高、低剂量组灌胃给予治疗。假手术组和脑外伤组给予相应体积的等渗生理盐水,采用平衡木方法评估4组脑外伤大鼠的神经功能恢复情况。各组大鼠血清通过酶联免疫吸附测定法,测定其TNF-α和IL-6含量,取各组大鼠脑组织,通过免疫组化法去测定神经元特异性烯醇化酶(NSE)表达情况,Western blot测定各组大鼠损伤侧脑皮层组织PI3K蛋白的表达情况。结果:损伤后的第6、24、60小时3个时间点提示,草决明提取物高、低剂量和脑外伤组的平衡功能受损恢复明显比假手术组低(P<0.01),但草决明提取物高、低剂量组的平衡功能受损恢复较脑外伤组快(P<0.05)。6,24,60 h 3个时间点脑外伤组和草决明提取物高、低剂量组的血清TNF-α和IL-6水平比假手术组明显增高(P<0.01),两给药组的大鼠血清TNF-α、IL-6水平低于脑外伤组(P<0.05),整体水平24 h逐渐下降,而60小时时略有反弹增高。与假手术组相比,两给药组的NSE阳性细胞表达明显降低(P<0.01),而明显高于脑外伤组(P<0.05);脑外伤组PI3K阳性表达明显低于假手术组(P<0.05),草决明提取物高剂量组PI3K阳性表达明显高于脑外伤组(P<0.05)且与假手术组持平。结论:草决明提取物能够减轻大鼠脑水肿,并增高脑外伤大鼠NSE阳性细胞表达,继而提高PI3K蛋白的表达水平从而加速脑损伤后神经功能的恢复。 Objective To assess the effect of Cassia o btusifolial extract on rats of with traumatic brain injury.MethodsIn total of 70 rats were randomly divided into 4 groups,the traumatic brain injury(TBI)group,the sham operation(SHAM)group,the(COB-H20 g/kg、COB-L10 g/kg)of Cassia extract group COB-H20 g/kg group,and the COB-L10 g/kg groups.All the other groups They were subjected to the modified Feeney's weight-drop model except for the SHAM operation group.The sham group operation group received fake open skull window treatment only,while the COB-H20 g/kg group and the COB-L10 g/kg group Cassia o btusifolial extract were given by intragastric administration and the rats were given isotonic salt of equal volume in the sham group and the traumatic brain injury group instead of TBI and SHAM.The recovery of nerve function in rats was evaluated by through Balance beam.TNF-αand IL-6 level were detected by ELASA method in serum on rats of TBI.The NSE positive cells near the region of injury was ascertained by measuring in rats of TBI,and Western blot was used to detect the expressions of PI3K in rat thebrain tissues.ResultsThe recovery of nerve function of Both of the COB-H20 g/kg group,the COB-L10 g/kg group Cassia extract groups and the TBI group are significantly lower than that of the SHAM group when three time points were suggested at 6 h,24 h and 60 h after injury(P<0.01),and the recovery of nerve function in the COB-H20 g/kg group and the COB-L10 g/kg group was significantly better than that of the TBI(P<0.05).The serum TNF-αand IL-6 of content inthe TBI group,the COB-H20 g/kg group,and the COB-L10 g/kg group were significant higher than that of the SHAM group(P<0.01).The serum TNF-αand IL-6 of the COB-H20 g/kg group and the COB-L10 g/kg group were significantly lower than that of the TBI group(P<0.05).And both of COB groups were significantly higher than sham group at 6,24,60h(P<0.01).To be compared with the TBI group,the serum TNF-αand IL-6 content in both COB groups significantly decreased respectively at the ti
